Purpose:The objective of this study is to examine the expression patterns of eif3b in both colorectal cancer cells and tissues,and evaluate its impact on cell proliferation and metastasis.Additionally,we aim to investigate the potential diagnostic and therapeutic value of eif3b as a biomarker and target for colorectal cancer patients.Methods:1.Expression of eif3b in colorectal cancer tissue samplesIn this study,immunohistochemistry was employed to detect the expression level of eif3b protein in colorectal cancer tissue slices and compare it with that in normal tissue.Subsequently,we analyzed the clinicopathological parameters of colorectal cancer patients,including age,gender,tumor size,degree of differentiation,and lymph node metastasis,to evaluate the correlation between the expression level of eif3b and these parameters.Our aim was to assess whether eif3b coμld serve as a potential diagnostic biomarker for colorectal cancer.2.Effects of eif3b on proliferation,invasion and migration of colorectal cancer cellsThe slow virus eif3b-sh RNA reagent was transfected into colorectal cancer cells,and then the effect of eif3b gene expression was detected using QRT-PCR technology.The effect of down-regμlating eif3b on the proliferation ability of colorectal cancer cells was evaluated by CCK8 cell proliferation experiment.The transwell migration assay was used to assess the impact of eif3b down-regμlation on the migratory ability of colorectal cancer cells.Similarly,the transwell invasion assay was performed to evaluate the effect of eif3b knockdown on the invasive potential of colorectal cancer cells.Resμlts:1.eif3b was highly expressed in colorectal cancer tissue samplesIn this study,immunohistochemistry was used to detect the expression level of eif3b protein in colorectal cancer tissue and its adjacent normal tissue.The resμlts showed that the protein expression level of eif3b was significantly higher in colorectal cancer tissue compared to its adjacent normal tissue.Further analysis revealed that eif3b was predominantly expressed in the cytoplasm of cells in colorectal cancer tissue.2.Down-regμlating eif3b expression inhibits proliferation,invasion and migration of colorectal cancer cellsIn this study,QRT-PCR was employed to detect the expression level of eif3b gene in Caco2 colorectal cancer cells that had been transfected with lentivirus eif3b-sh RNA reagent.The experimental resμlts showed that compared to the control group,both eif3b m RNA and protein levels were significantly decreased in the experimental group,indicating that eif3b-sh RNA reagent effectively down-regμlated the expression of eif3b in Caco2 colorectal cancer cells.To evaluate the effects of down-regμlating eif3b on colorectal cancer cells,we conducted a series of in vitro cell experiments.According to the findings of the transwell invasion assay,the invasive ability of Caco2 colorectal cancer cells was considerably suppressed upon down-regμlation of eif3b expression.Similarly,the resμlts of the transwell migration assay indicate that the migratory capability of Caco2 colorectal cancer cells was significantly impeded upon eif3b down-regμlation.Conclusion:1.The expression level of eif3b is known to be upregμlated in human colorectal cancer tissue,and has been found to be positively correlated with tumor stage,degree of differentiation,and lymph node metastasis.These findings sμggest that eif3b may serve as a promising new diagnostic biomarker for colorectal cancer。2.Eif3b can promote the proliferation,invasion and migration of colorectal cancer cells。...
