Objective:A model of bilateral renal ischemia-reperfusion acute kidney injury in mice was established,and the protective effect of short-chain fatty acids on mouse kidneys was observed by detecting renal function and renal pathology,and the expression levels of tumor ne Crosis factor-α(TNF-α),CD68 and Toll-like receptor 4(TLR4)proteins in kidney tissues were further detected.To investigate the inhibitory effect of short-chain fatty acids on renal inflammation after renal ischemia-reperfusion injury in mice.Methods:Fifteen mice were randomly divided into three groups:sham group,IRI group,IRI+SCFAs group,5 rats in each group.Mice in IRI+SCFAs group were fed with short-chain fatty acids for 7 days before operation.Serum and kidney samples were collected 48 h after ischemia-reperfusion,serum Creatinine and urea nitrogen were detected.Renal tissue was stained with hematoxylin eosin(HE),and TNF-α and CD68 were detected by immunohistochemistry(IHC).The expression of TLR4 protein was detected by Western blot(WB).Results:1.The levels of Creatinine and urea nitrogen in IRI group were significantly higher than those in sham group(p<0.05).The levels of Creatinine and urea nitrogen in IRI+SCFAs group were significantly lower than those in IRI group(p<0.05).2.In IRI group,renal tissue structure was damaged,vacuolar degeneration and ne Crosis of renal tubular epithelial cells,brush border exfoliation,renal tubular cast and a large number of inflammatory cell infiltration were observed in renal interstitium,and the pathological score of injury was significantly higher than that of sham group(p<0.05).The pathological injury and inflammatory cell infiltration of renal tissue in IRI+SCFAs group were significantly less than those in IRI group(p<0.05).3.Immunohistochemical staining showed that there was no positive expression of TNF-α and CD68 in renal tissue of sham group.The expression of TNF-α and CD68 in renal tissue of IRI group was significantly higher than that of sham group(p<0.05).In IRI+SCFAs group,TNF-α and CD68 were only expressed in a small amount in renal tissue,and the semiquantitative score was statistically significant(p<0.05).4.The expression level of TLR4 protein in IRI group was significantly higher than that in sham group(p<0.05).The expression level of TLR4 protein in IRI+SCFAs group was significantly lower than that in IRI group(p<0.05).Conclusion:1.Exogenous administration of short-chain fatty acids can alleviate renal injury after ischemia-reperfusion in mice.2.Short-chain fatty acids may inhibit renal inflammatory response by influencing TLR4 expression,and ultimately exert a protective effect on the kidneys.
