| Objective:To observe the effect of adiponectin receptor agonist(AdipoRon)on the expression of endoplasmic reticulum stress in renal tissue of streptozotocin-induced diabetic mice.Methods:Thirty 7-week-old male C57 BL / 6 mice were randomly divided into normal control group(n = 6)and diabetic model group(n = 24).The type 1 diabetic mouse model was prepared by intraperitoneal injection of streptozotocin(60 mg / kg)for 5 days.One week later,the blood glucose was greater than or equal to 16.7 mmol / L,which was considered as successful modeling.A total of 18 mice were successfully modeled.The model mice were divided into two groups by random number table method,with 9 mice in each group,which were diabetic group(n = 9)and AdipoRon treatment group(n =9).The normal control group and the diabetic group were given the same amount of normal saline by gavage,and the AdipoRon treatment group was given AdipoRon(20mg / kg · d)by gavage.After 8 weeks of drug intervention,10 % chloral hydrate was injected intraperitoneally for anesthesia,and blood samples were collected from the inner canthus vein for blood glucose,insulin,urea nitrogen and creatinine determination.After blood collection,the animals were sacrificed immediately,and the kidneys on both sides were quickly removed.The kidneys were washed in normal saline and the water was removed by filter paper.After weighing,the ratio of kidney weight to body weight was calculated.Part of the kidney tissue was fixed with 10 % neutral formalin solution for HE staining.The remaining part of the kidney tissue was immediately placed in liquid nitrogen and stored in a-80 ° refrigerator for the detection of endoplasmic reticulum stress indicators(GRP78,CHOP,Caspase-12)mRNA.During the experiment,all mice in each group died,and 6 mice remained in each group after the experiment.Results:Compared with the normal control group,the levels of blood glucose,creatinine,urea nitrogen and kidney weight/body weight in the diabetic model group were significantly increased(P<0.05),the insulin level was significantly decreased,the renal pathological changes were aggravated,and the expression of GRP78,CHOP and Caspase-12 mRNA in renal tissue was significantly increased(P<0.05).Compared with the diabetic model group,the levels of blood glucose,creatinine,urea nitrogen and kidney weight/body weight in the diabetic adiponectin receptor agonist group were significantly decreased(P<0.05).Insulin level was significantly restored(P<0.05),renal pathological changes were alleviated,and the expression levels of GRP78,CHOP and Caspase-12 mRNA in kidney were significantly decreased(P<0.05).Conclusion:The blood glucose level of the diabetic model group increased,the insulin level decreased,the renal function decreased,and the renal endoplasmic reticulum stress mRNA level increased.After AdipoRon intervention,the blood glucose level of the diabetic mice decreased,the insulin level increased,the renal function improved,and the endoplasmic reticulum stress mRNA expression level decreased,suggesting that Adi Po Ron may play a protective role in the kidney of diabetic mice by regulating endoplasmic reticulum stress. |
