| BACKGROUND:According to the GLOBOCAN 2020 estimates,Gastric cancer ranks as the fourth leading cause of cancer deaths in both genders combined.One of the main predisposing causes of gastric cancer is Helicobacter pylori(H.pylori,Hp).There is significantly increasing about the effect of Hp on the progression of gastric cancer since Meyer and others discovered the possible mechanism of Hp inducing gastric cancer in 2017.The focus of current gastric cancer research is the relationship between Hp and energy metabolism of gastric cancer cells.Hp infection can triggers oxidative stress reaction in the body.In order to maintain cell growth and proliferation,studies shows that tumor cells acquire large amounts of energy through aerobic glycolysis while increasing their antioxidant capacity.Hp infection is closely related to glycolytic energy metabolism by triggering oxidative stress reactions in the body.JMJD5(Jumonji C domain 5)is a dioxygenase containing Jumonji C domain.The expression of JMJD5 will be up-regulated under the condition of hypoxia,and it can promote tumor cells to proceed along the direction of glycolysis by regulating the key enzymes in the glycometabolism pathway of cancer cells,so as to generate more energy to support the growth and proliferation of tumor cells.It is speculated that the expression of JMJD5 may be affected by Hp infection and through their interaction regulating glycolysis.This study detected the expression of JMJD5 and Hp infection in gastric cancer tissue,and further explored the relationship between JMJD5 and Hp in gastric cancer tissue,the study will offering theoretical basis for early phase diagnosis,prognosis evaluation,and curing target exploration of gastric cancer.OBJECTIVE:Through detect the expression of JMJD5 and Hp infection in gastric cancer and chronic non-atrophic gastritis,to explore the role of JMJD5 and Hp in the development of gastric cancer.METHODS:From September 2021 to December 2022,93 cases of newly diagnosed and surgically resected tissue samples of gastric cancer from the Department of Gastroenterology and General Surgery of Huaihe Hospital of Henan University were collected;93 tissue samples of chronic non-atrophic gastritis from the same term were selected from this hospital through endoscopy and gastric mucosal biopsy.Two methods of13C breath test and immunohistochemical test were used for the diagnosis of Hp infection in the mucosa of the two tissues.The results of the two detection methods were both positive to determine Hp positive.If the two test methods are one Yin and one Yang or both are negative,it is judged as Hp negative.According to the difference of Helicobacter pylori infection,each group was divided into Hp positive group(Hp+)and Hp negative group(Hp-).The expression of JMJD5 protein in Hp+and Hp-was tested by immunohistochemistry.The relationship between Hp,JMJD5 and clinicopathological data(age,sex,tumor size,depth of invasion,degree of the tumor differentiation,TNM stage,lymph node metastasis,CEA,CA125,CA199)of patients with gastric cancer was statistically analyzed,as well as the correlation between JMJD5 and Hp infection.RESULTS:1.Hp infection in different gastric mucosa tissues:group of gastric cancer(79.6%)>group of chronic non-atrophic gastritis(48.4%),there is significant differences between the two groups(χ2=19.619,P<0.001).2.Hp infection in gastric cancer samples had no significant differences with gender,age,tumor size,CEA,CA125,CA199(P>0.05),but correlated to and show significant differences with depth of tumor invasion,degree of differentiation,TNM stage and lymph node metastasis(P<0.05).3.The expression of JMJD5 in different gastric mucosa tissues:group of gastric cancer(63.4%)>group of chronic non-atrophic gastritis(37.6%),there is significant differences between the two groups(χ2=12.389,P<0.001).4.The expression of JMJD5 in gastric cancer had no significant differences with gender,age,tumor size,CA125,CA199(P>0.05),but correlated to and show significant differences with depth of tumor invasion,degree of differentiation,TNM stage,lymph node metastasis and CEA(P<0.05).5.Relationship between JMJD5 expression and Hp infection:The expression of JMJD5 in HP-positive group was significantly higher than that in HP-negative group(P=0.030),and there was a significant positive correlation between Hp infection and JMJD5 expression in gastric cancer tissues(χ2=4.687,r=0.224,P=0.030).CONCLUSION:1.Hp infection and JMJD5 protein expression are both related to the depth of tumor invasion,degree of differentiation,TNM stage,and lymph node metastasis of gastric cancer.2.There is a positive correlation between Hp infection and JMJD5 expression in gastric cancer tissues.Hp infection may participate in glycolytic metabolism of gastric cancer cells through up-regulation of JMJD5 expression. |
