Lung cancer is a malignant tumor with high morbidity and mortality worldwide,which poses a serious threat to human health.Lung adenocarcinoma is the largest subtype of all pathological types of lung cancer and has the highest incidence.The in-depth study of the core genes of lung adenocarcinoma is of great significance for the early treatment and prognosis of lung adenocarcinoma.In this paper,the network analysis method is used to analyze the gene expression profile data of lung adenocarcinoma,and finally the key genes significantly related to the clinical characteristics and survival status of lung adenocarcinoma patients are obtained,and the key genes are analyzed and verified.Based on the RNA-seq gene expression profile data of 598 lung adenocarcinoma samples from the TCGA database,this paper first screens out differentially expressed genes.Through enrichment analysis,the differentially expressed genes are mainly involved in immune-related pathways,immune-related diseases,cellular processes and many metabolic pathways.The protein interaction network was constructed by using differential genes.Through cluster analysis,the network with the most compact nodes was obtained.The closeness centrality,betweenness centrality and degree centrality values of each gene in the network were calculated.The top 43 genes were screened out,and 14 key genes were obtained by intersection.Secondly,a weighted gene co-expression network was constructed using cancer-normal sample data,and 25 gene modules were obtained by cluster analysis,and 394 key genes of 7 modules significantly related to cancer status were screened out.Thirdly,the weighted gene co-expression network was constructed by using cancer sample data,and the gene module was combined with clinical information.By analyzing the correlation between the module and the patient’s age,gender,smoking age,cancer stage and survival status,51 key genes in key module were screened.Finally,the gene expression data of lung adenocarcinoma were analyzed by combining protein interaction network and weighted gene co-expression network.The obtained key genes were intersected and six key genes were finally determined: CDC20,NCAPG,RRM2,CENPA,HJURP and KIF2 C.In this paper,the analysis of gene expression showed that the expression of key genes was highly expressed in lung adenocarcinoma,and increased with the increase of tumor stage.The prognosis of key genes was significantly different,and the survival rate of high-risk samples was lower than that of these low-risk samples.Through keyword co-occurrence network analysis,it was found that key genes were highly correlated with lung adenocarcinoma and other cancer research.Therefore,the research method of this paper provides a reference for the screening of key genes in lung adenocarcinoma.Six key genes are closely related to the occurrence and development of lung adenocarcinoma,and provide suggestions and help for the diagnosis and treatment of lung adenocarcinoma.
