| The metabolites of two Marine Aspergillus fungi were studied in this paper,including two topics.(1)Structural analysis and activity study of polyketones,alkaloids and other components in Aspergillus fumigatus CBC18132 secondary metabolites.(2)Chiral resolution of aromatic myrrh gefinoid compounds metabolized by Aspergillus versicolor YPH93,studies of similar structures in the literature,and the discovery of a natural iron death inhibitor.The structures and activities of important species of Aspergillus fungi of Marine origin from 2014 to 2021 are reviewed.Aspergillus fungi of Marine origin can produce compounds with novel structures and significant biological activities.The secondary metabolites of A.fumigatus CBC18132 were studied.The secondary metabolites of A.fumigatus CBC18132 were isolated by MCI resin,ODS,silica gel column chromatography,semipreparative HPLC and other chromatographic chromatography techniques based on the spectral and mass spectral analysis of crude extracts.The structure of the compound was identified using nuclear magnetic resonance spectroscopy(1H NMR,13 C NMR,2D NNR),high-resolution mass spectrometry(HRESIMS),ultraviolet spectroscopy(UV),and circular dichroism spectroscopy(CD).Twenty-two compounds were isolated from the strain,including two new polyketones(1-2),six known polyketones(3-7,22),and 14 known alkaloids(8-21)(mainly indoles and quinazolines).The aromatic sesquiterpenes metabolized by A.versicolor YPH93 were studied.We have obtained 17 aromatic myrrh sesquiterpenes from this strain,including 7 new aromatic myrrh sesquiterpenes(23-26,28,29,31)and 10 biogenic derivatives(27,30,32-39),but have not confirmed their absolute configuration or screened their activity.Three pairs of enantiomers were obtained by chiral resolution.Compounds 23-28 and 30-32 were derivatives of sydowic acid.By analyzing these structures in literature,it was found that some compounds had similar structures and circular dichroic data,but C-7 configurations were different.There is misdetermination.In this paper,the two common conformations of pyranoid rings with this structure,the relationship between the chirality of C-7 and the Cotton effect at 210 nm in CD spectra,and the structures of 6 compounds reported in the literature(mainly absolute configurations)were studied in depth.The activity of some metabolites of two strains of fungi was studied.Evaluation of compound antioxidant activity,α-glycosidase activity,cytotoxic activity of enzyme inhibition,iron activity of death.Compound 4 was found to induce erastin/ RSL3-induced iron death in HT1080 cells at 10 μM.Compound 29 showed moderate inhibition of erastin/ RSL3-induced iron death,with EC50 values ranging from 2 to 4 μM,and compound 30 also showed weak inhibition of iron death(about 50%)at 10 μM.In the first part of this paper,the secondary metabolites of A.fumigatus CBC18132 were studied.Two new compounds were found and the absolute configuration of compound 4 was determined for the first time,which enriched the metabolites of Aspergillus fungi.In the second part,the structure and activity of 17 aromatic sesquiterpenoids were studied,the relationship between the spectral data and the configuration of these compounds was summarized,which provided a theoretical basis for the structure determination of these compounds,and one iron death inhibitor was found. |
