| Ganoderma lucidum is a kind of poraceae fungus.Having the effect of lowering blood sugar,lowering blood fat,improving immune function and fighting aging.The alcohol extract of Ganoderma lucidum has good antioxidant capacity,which can increase the activity of antioxidant enzyme and reduce the oxidative free radicals in cells.Insulin is the only hypoglycemic hormone in human body,and its physiological function is mainly to promote the synthesis of glycogen in liver and muscle tissue,promote the synthesis of fat and protein and inhibit their decomposition.Insulin resistance can cause diabetes.After insulin resistance,the body’s sensitivity to insulin decreases and the body promotes a decrease in glucose intake.When the body has insulin resistance,insulin in adipose cells cannot well inhibit lipolysis,the rate of lipolysis of adipose cells increases,and the level of free fatty acids in plasma increases to form hyperlipidemia.Meanwhile,free fatty acids ectopically accumulate in other tissues,further causing insulin resistance in peripheral tissues and the whole body.Research on inhibiting insulin of adipocyte is beneficial to control insulin resistance from the source and alleviate the occurrence of diabetes.Oxidative stress plays a key role in the development and intensification of diabetes mellitus and its complications,and antioxidant therapy can inhibit or delay the development of diabetes mellitus and chronic complications.In this paper,by studying the effect of Ganoderma lucidum alcohol extract on insulin resistance of adipocytes,the pharmacological of improving insulin resistance of Ganoderma lucidum alcohol extract was explored.The main research results are as follows:(1)Adipocytes were treated with 1μmol/ l dexamethasone for 72 h to establish insulin resistance and oxidative stress models.(2)Glucose consumption was detected by glucose oxidase method,triglyceride was detected by GPO-PAP method,ROS was detected by DCFDA fluorescence probe method,ROS fluorescence value and mitochondrial membrane potential were detected by JC-1fluorescence staining,and the effects of Ganoderma lucidum alcohol extract on insulin resistance and oxidative stress of adipocytes were analyzed.The results showed that Ganoderma lucidum ethanol extract could significantly increase glucose consumption and intracellular triglyceride content in a dose-dependent manner.The alcohol extract of Ganoderma lucidum can significantly reduce ROS content and mitochondrial membrane damage of adipocytes.(3)Western blot was used to detect the proteins related to oxidative stress and insulin signaling pathway.After the alcohol extract of Ganoderma lucidum was treated,the expression of NOX4 and PTP1 B proteins was down-regulated,indicating that the oxidative stress pathway was inhibited.The alcohol extract of Ganoderma lucidum promoted the expression of IRS1,PI3Kp110,PI3Kp85,p-AKT,p-GSK3β and Gl UT4 proteins,indicating that insulin signaling pathway was activated.The results showed that 10 μg/ml alcohol extract of Ganoderma lucidum could significantly improve the oxidative stress and insulin resistance of adipocytes by down-regulating NOX4 expression to reduce the ROS production of adipocytes,further inhibit the expression of PTP1 B protein and activate the IRS1-PI3K-AKT insulin signaling pathway.The expression of IRS1,PI3 K,p-AKT,p-GSK3β and Gl UT4 can improve the insulin resistance of adipocytes by promoting the glucose uptake ability of adipocytes and increasing the synthesis of triglycerides in adipocytes. |
