Since fermented Cordyceps powder Cs-4 contains a large amount of nutrients and is susceptible to microbial contamination,the microbial counts of the samples sterilized by the existing sterilization methods are easily exceeded during the production,storage and transportation at room temperature.In order to ensure the hygienic quality of drugs,the sterilization treatment of fermented Cordyceps powder needs to be replaced,and 60Co-γray irradiation sterilization is the sterilization method recognized by the Chinese Pharmacopoeia.In this paper,fermented Cordyceps powder before and after 60Co-γray irradiation sterilization was used as the research object,and the irradiation sterilization process parameters were investigated with reference to the provisions under the content determination of Jin Shui Bao capsule in the 2020 edition of Chinese Pharmacopoeia,with nucleoside(uridine,guanosine,adenosine)and sterol(ergosterol),and the content of uracil and adenine in nucleoside were added as indicators.In order to scientifically and comprehensively evaluate the quality difference of fermented Cordyceps powder before and after irradiation sterilization,on the basis of the indexes for measuring the content of nucleoside(uridine,guanosine,adenosine)and sterol(ergosterol)contained in the Chinese Pharmacopoeia 2020 edition,the detection methods for polysaccharide,monosaccharide,fatty acids,sugar alcohol,amino acids and other components in fermented Cordyceps powder are added,which can comprehensively analyze and evaluate the quality difference of fermented Cordyceps powder before and after irradiation,and provide a basis for the feasibility of applying 60Co-γray irradiation sterilization to fermented Cordyceps powder,and the main research content is divided into three parts.(1)Investigation for irradiation sterilization process parameters of fermented Cordyceps powder Cs-4:By measuring the content of five nucleoside and sterol in fermented Cordyceps powder at three different irradiation positions and five different irradiation doses of 4,6,8,10 and 12 k Gy,and combining the results of microbial detection and long-term toxicity test,we investigated the effects of irradiation mode,irradiation dose,irradiation frequency and irradiation time on irradiation process,and selected the most reasonable irradiation sterilization position of fermented Cordyceps powder as middle 4 positions and irradiation sterilization dose as 8 k Gy.(2)Establishment of multi-indicator component assay method for fermented Cordyceps powder Cs-4:The method for determination of total polysaccharide,4monosaccharide,5 fatty acids and mannitol in fermented Cordyceps powder and HPLC fingerprint study method for 17 amino acids were added.The method is simple,reliable,reproducible and can accurately determine several chemical components in fermented Cordyceps powder.It is suitable for quality assessment and control of fermented Cordyceps powder.(3)Stability study of fermented Cordyceps powder Cs-4 before and after irradiation sterilization:With reference to the 2020 edition of the Chinese Pharmacopoeia 9001,five nucleoside,ergosterol,total polysaccharide,four monosaccharide,five fatty acid,mannitol and 17 amino acid in fermented Cordyceps powder before and after 60Co-γray radiation sterilization were investigated in the influencing factor test,accelerated test and long-term test.The results showed that there was no significant effect of light,high temperature and high humidity on the content of each component in fermented Cordyceps powder after irradiation sterilization,the quality of each index component was relatively stable under the accelerated and long-term test conditions after the sample was irradiated and sterilized,there was no significant effect of each component in the sample before and after irradiation sterilization,and HPLC fingerprint similarity of 17 amino acid was above 0.95,which better ensured the quality fluctuation and change of the fermented Cordyceps powder before and after irradiation sterilization.In this paper,through the selection of process parameters,the establishment of index component detection method and study of stability before and after irradiation sterilization,the most reasonable irradiation sterilization process parameters were selected for the application of 60Co-γray irradiation to fermented Cordyceps powder,which can provide accurate and fast content determination and fingerprint detection method for each component in fermented Cordyceps powder,and provide a basis for the feasibility of applying 60Co-γray irradiation sterilization to fermented Cordyceps powder. |
