| Research Objective: This project aims to explore the mechanism of action of steamed umbilical cord therapy to improve visceral hypersensitivity in the treatment of IBS-D by regulating substance P in cerebroenteric peptides,provide more simple,convenient,effective and inexpensive treatment methods for IBS-D patients,and also provide new ideas and basis for the clinical treatment of IBS-D.Research Methods: In this project,40 male rats with SD were selected and divided into blank group,model group,steamed umbilical group,and western medicine control group,with 10 rats in each group,and single-cage feeding.Except for the blank group,the rats in the other groups were modeled: senna leachate was given gavage,combined with chronic mild restraint stress and solitary feeding method to stimulate the rats.Following successful modelling,the blank and model group were filled with 10 ml/kg of standard saline solution daily.The western drug control group was given pivylomamine suspension for gavage at a concentration of 3 mg/m L.After steaming the umbilical cord,the drug end was fixed at the rat Shenque point with a non-woven wrapped bundle,and the elasticity was suitable,and it was removed after retaining it for24 h.1 time every other day.After the above three groups of treatment methods were continuously intervened for 14 years,materials and sample preparation were taken to evaluate the intestinal sensitivity of rats before and after treatment,the serum and SP content of colon and hippocampal tissues of rats were determined by ELISA method,the pathological observation of colon tissues was performed,and finally statistical software was used to statistically analyze the data,and the mechanism of action of steamed umbilical cord therapy to improve visceral hypersensitivity in the treatment of IBS-D by regulating P substance was discussed according to the results.Research results:General condition of rats in each group: before the start of modeling,there were no obvious differences in mental state,defecation status,fecal traits,eating and drinking water of rats in each group;After the end of modeling,the stool traits of the rats in the control group were basically lumpy or sausage-like,with normal appetite,flexible movements,full of energy,white and shiny coat,while rats in the model group,steamed navel group and western medicine control group all had different degrees of diarrhea,Bristol score was significantly increased,and the difference was statistically significant(P<0.05),manifested as pasty,muddy or watery stool,mental burnout,reduced activity,owed coat,and reduced eating.After the end of the drug intervention,compared with the blank control group,the rats in the model group had severe diarrhea,higher Bristol score(P<0.05),yellow coat,but normal spirit,activity and eating.Compared with the model group,the degree of diarrhea in the western medicine control group and the Chinese medicine steamed navel group was reduced,and the Bristol score was lower than that in the model group(P<0.05),with normal spirit and activity,shiny coat,but reduced eating.Compared with the western medicine control group,the diarrhea degree of rats in the steamed navel group was significantly improved(P<0.05),their spirit,activity and eating were normal,and the coat was shiny.There were no deaths in the experiment.It suggested that the above symptoms of rats in the steamed navel group and the western medicine group were improved after drug intervention,but the efficacy of the steamed navel group was more significant.2.Changes in body mass of rats before,after and after treatment in each group: there was no significant difference in body mass between groups before the start of modeling(P>0.05);After the completion of molding,compared with the blank group,the body weight of the modeled rats(model group,western medicine group,steamed umbilical group)was significantly reduced,and there was a significant difference(P<0.01).After the drug intervention,compared with the blank group,the average body mass of rats in the model group,western medicine control group and Chinese medicine steamed navel group was significantly reduced,and the difference was statistically significant(P<0.01).Compared with the model group,the body mass of rats in each dosing group increased(P<0.05),and the average body weight of rats in the steamed umbilical group was higher(P<0.05).3.The proportion of dry and wet feces in rats before and after treatment: before treatment,compared with the blank group,the proportion of dry and wet feces in the model group,western medicine group and steamed navel group was significantly increased(P<0.01);After treatment,compared with the model group,the proportion of dry and wet feces in each administration group decreased(P<0.01),and the proportion of dry and wet feces in the Chinese medicine steamed navel group was more significant(P<0.01).Conclusion: Umbilical steaming therapy can significantly improve the general condition of IBS-D model rats,reduce the content of SP in hippocampal tissue,serum and colon tissue,regulate visceral hypersensitivity,and reduce intestinal inflammation,thereby alleviating the clinical symptoms of IBS-D. |
