| Objectives1.to determine the expression of Annexin A10(ANXA10)in Hilar cholangiocarcinoma(HCCA)tissues and paracancerous tissues by Immunohistochemistry(IHC).2.detection of serum levels of Carbohydrate antigen 19-9(CA19-9)in HCCA patients and patients with common bile duct stones.3.to examine the extent of ANXA10 expression and CA19-9 levels in HCCA tissues and the relationship between clinicopathological factors(i.e.,age,gender,stage of tumor diameter differentiation,lymphatic metastases,vascular nerve infiltration)and prognosis(survival time)of patients.4.to investigate the role and relevance of ANXA10 expression and CA19-9 levels in the development and progression of HCCA.Methods1.clinical data and pathological specimens were collected from 40 patients who attended the Department of Hepatobiliary,Pancreatic and Splenic Surgery at xxx Hospital for radical surgery for HCCA between January 2013 and December 2021,as well as clinically relevant data from 40 patients with common bile duct stones treated surgically during the same period.the experimental group for ANXA10 was the selected 40 cases of HCCA cancer tissue,and the control group was paracancerous tissue(determined by The experimental group for the detection of CA19-9 was the selected 40 patients with HCCA,and the control group was the40 patients with common bile duct stones.2.The expression of ANXA10 in HCCA tissues and paracancerous tissues was determined by IHC.CA19-9 levels of HCCA patients were measured and grouped according to their clinical information,including age,gender,tumor diameter,differentiation,stage,lymphatic metastasis,and vascular and nerve infiltration.The degree of ANXA10 expression and preoperative CA19-9 levels were analyzed in correlation with various factors and their role in HCCA.3.To analyze the variability of ANXA10 expression in cancer and paraneoplastic tissues of HCCA patients and the variability of CA19-9 expression levels with those of patients with common bile duct stones by SPSS 22.0 statistical software,and to explore the correlation between the upper two and clinical factors by using the chi-square test to investigate the correlation between ANXA10 expression levels and preoperative CA19-9 levels in HCCA patients using SPEARMAN rank correlation analysis.A P value < 0.05 was considered statistically significant.4.The Kaplane-Meier method and log-rank method allowed us to obtain survival curves of HCCA patients,as well as variables associated with prognosis,and to include variables with P<0.05 in the COX stepwise regression model to explore their association with forecast and ultimately to identify independent risk factors affecting the prognosis of HCCA patients.Results1.This study found a significant difference in ANXA10 expression levels between HCCA cancer and paraneoplastic tissues,with a statistical analysis of correlation of P= 0.00018.by statistical study,ANXA10 expression was significantly correlated with tumor ≥ 2.5 cm,hypofractionation,T3\T4 stage,lymphatic metastasis(+),and vascular infiltration(+)positive correlation(P<0.05).at the same time,there was no association with patient age,gender,and neuro infiltration(P>0.05).2.The results showed that there was a significant difference in CA19-9 levels between patients with HCCA and patients with common bile duct stones(P<0.05).According to statistical analysis,there was a significant correlation between CA19-9 levels in HCCA patients and their age,degree of differentiation,tumor stage,lymphatic metastasis,and vascular infiltration(P<0.05).In contrast,there was no significant correlation with patients’ gender,tumor diameter,and nerve infiltration(P>0.05).3.As shown by SPEARMAN grade correlation analysis,ANXA10 expression was positively correlated with serum CA19-9 level in HCCA patients with a correlation coefficient R=0.899(P<0.05),which indicated a synergistic effect between ANXA10 and CA19-9 in HCCA patients.4.After univariate statistical analysis,we found that a significant increase in ANXA10,a considerable increase in CA19-9,age 60 years or older,lymphatic metastasis(+),T3/T4 stage,and vascular infiltration(+)were statistically significant in correlating with poor postoperative prognosis in HCCA patients.Multifactorial analysis showed that lymphatic metastasis(+)and vascular infiltration(+),and age ≥ 60 years were statistically significant independent risk factors for prognosis in patients with HCCA.Conclusions1.The results showed that ANXA10 expression was higher in HCCA cancer tissues than in paracancerous tissues,indicating that ANXA10 influences HCCA progression.2.CA19-9 levels were higher in HCCA patients than in patients with common bile duct stones,suggesting that CA19-9 affects HCCA progression.3.High expression of ANXA10 in HCCA and high serum CA19-9 levels were strongly associated with T3\T4 stage,lymphatic metastasis(+),and vascular infiltration(+),and both had a synergistic effect in HCCA patients;lymphatic metastasis(+)and vascular infiltration(+),and age ≥ 60 years,were factors affecting the prognosis of HCCA patients,which suggests that they have a poor prognosis and short survival.4.In the future,a combined test for ANXA10 and CA19-9 may help in the diagnosis of HCCA;further studies on ANXA10 may provide new targets for later targeted therapies. |
