| Objective To investigate the expression of ribosomal protein L31(RPL31)in colorectal cancer and normal colorectal tissues,and to study the correlation between the clinicopathological features and the expression of RPL31,and to analyze whether the expression of RPL31 can predict the prognosis of patients with colorectal cancer.Methods(1)Human normal colorectal cell line FHC and colorectal cancer cell lines HCT 116,RKO,HT29 were cultured in vitro.The expression abundance of RPL31 gene in each cell was detected by q RT-PCR.(2)Tissue microarray(including 101 colorectal cancer tissues and77 normal tissues adjacent to cancer)was purchased from Shanghai Aoduo Biotechnology Co.,Ltd.,and the expression of RPL 31 was determined by immunohistochemistry.The relationship between the expression of RPL31 and clinicopathological features in colorectal cancer was further studied.(3)The survival time of the patients was calculated and the survival curve was made for survival analysis.Results(1)q RT-PCR showed that RPL31 m RNA was highly expressed in CRC cell lines,especially in HCT 116 and RKO cells,and the difference was statistically significant(P<0.05).(2)Immunohistochemical results showed that the expression rate of RPL31 in colorectal cancer tissues was 50.5%(51/101),which was higher than that in adjacent tissues(6.5%(5/77),P<0.05).(3)The expression level of RPL31 was significantly correlated with tumor size(P<0.05),but not with sex,TNM stage,pathological grade and lymph node metastasis(P>0.05).(4)The expression of RPL 31 gene was significantly correlated with the overall survival of colorectal cancer.Conclusion The expression of RPL31 is higher in colorectal cancer tissues than that in adjacent tissues,and the expression level of RPL31 is correlated with tumor size,that is,the expression of RPL31 increases with the increase of tumor size.In addition,the expression of RPL31 was correlated with the overall survival of colorectal cancer.With the increase of RPL31 gene expression,the survival of patients was shortened.RPL31 may be a potential oncogenic factor in colorectal cancer.The expression of RPL31 is helpful to reveal its role in the development of colorectal cancer and provide new ideas for clinical treatment and prognosis of colorectal cancer. |
