Objective:In recent years,the incidence rate and mortality of cancer in China have also been increasing.Lung cancer has become the fastest-growing malignant tumor in terms of incidence rate and mortality at home and abroad.At present,the treatment of lung cancer mainly depends on surgery,radiotherapy,and chemotherapy.Radiotherapy and chemotherapy can cause serious side effects.Therefore,it is urgent to seek new effective,targeted,and immune anti-tumor therapy.Many studies have shown that flavonoids,natural antioxidants with significant antioxidant activity,have shown good anti-tumor activity.As a member of flavonoids,myricetin has not been proved in detailed research on its antioxidant and anti-tumor activities.Based on this,this article focuses on the antioxidant and anti-tumor activities of myricetin and explores the possible biological mechanism of the anti-tumor effect of myricetin-Cu2+system,so as to lay a theoretical basis for future anti-cancer treatment.Methods:1、The antioxidant capacity of myricetin was analyzed to scavenge ABTS·+,DPPH radical,superoxide anion radical,hydroxyl radical and reduce Cu2+and Fe3+,as well as the in vitro protection damage of myricetin.2、MTT method and Western Blot technology were used to analyze the proliferation inhibitory effect of myricetin on lung cancer A549 cells.3、The full-wavelength spectra of myricetin-Cu2+reaction system under different p H conditions were determined.To detect the content of superoxide radicals,consumption of GSH,and degradation of DNA and proteins produced by the myricetin-Cu2+reaction system under different conditions.Results:1、Myricetin and vitamin C have similar ability to reduce transition metals Cu2+and Fe3+and scavenge free radicals;As well as the protective effect of damaged PC12 cells,it can inhibit the production of reactive oxygen species in cells.2、Myricetin can significantly inhibit the activity of lung cancer A549 cells,and the effect of myricetin in inhibiting the activity of lung cancer A549 cells can be antagonized by Cu2+complexing agent,and the apoptosis of myricetin inducing apoptosis of lung cancer A549 cells requires Cu2+mediation.3、The myricetin-Cu2+system can produce ROS and directly damage DNA and proteins,inducing apoptosis in lung cancer A549 cells.In addition,it can also indirectly induce apoptosis of lung cancer A549 cells through the consumption of GSH.Conclusions:Myricetin has significant antioxidant capacity;The myricetin-Cu2+system can produce a large amount of ROS,which in turn oxidizes DNA and proteins,and consumes GSH,which may be the main biological mechanism of the pro-oxidative activity of the myricetin-Cu2+system mediated by the anti-lung cancer.
