Objective:Gastric cancer,as one of the most common malignant tumors in the digestive tract in the clinical,is an important health problem worldwide.Studies in recent years have shown that Long non-coding RNA(lncRNA)plays a significant role in cancer biology,which is involved in the occurrence,metastasis and chemotherapy resistance of gastric cancer,and can be used as diagnostic,predictive and prognostic markers and new therapeutic targets for gastric cancer,with broad clinical application prospects.A new lncRNA AC126763.1,which has not been widely studied,was screened by bioinformatics methods and its expression level in gastric cancer and adjacent normal tissues was detected,so as to explore the role of lncRNA AC126763.1 in the proliferation,migration and invasion of gastric cancer cells.Methods: Using TCGA complete transcriptome data,we first screened differentially expressed lncRNA,mi RNA and m RNA,and then established the ce RNA regulatory network map of lncRNA and m RNA competitively binding mi RNA with reference to existing databases to screen out lncRNA AC126763.1,which is probably associated with the occurrence and development of gastric cancer.And reveal its potential mechanism of action in gastric cancer.Twenty pairs of cancer tissues and adjacent normal tissues of gastric cancer patients undergoing surgery in the Gastrointestinal surgery Department of Inner Mongolia Autonomous Region People’s Hospital(Hohhot,China)from December 2021 to October 2022 were collected.q RT-PCR was used to detect the expression of lncRNA AC126763.1 in the tissues and cell lines of gastric cancer.Silencing lncRNA AC126763.1 in AGS through lentivirus infection to construct a stable cell line.The ability of cell proliferation was evaluated by CCK-8 assay and clonogenesis assay.Scratch healing test and cell migration test were used to evaluate migration and invasion ability of the cells.SPSS 22.0 software is used to analyze all data.3 replicates are used in per independent trial.Data are expressed as mean ± standard deviation.The t-test was used to analyze the differences between the two groups.P <0.05 was statistically significant.Results:1.lncRNA AC126763.1 was overexpressed in TCGA-GC dataset.2.Compared with the adjacent normal tissues,lncRNA AC126763.1 was significantly increased in gastric cancer tissues.Silencing lncRNA AC126763.1significantly inhibited the proliferation,clonal formation,invasion and metastasis of human gastric adenocarcinoma cells(AGS).Conclusions:1.lncRNA AC126763.1 was overexpressed in cells and tissues of gastric cancer.2.lncRNA AC126763.1 can promote the growth,migration and invasion of gastric cancer cells,suggesting that lncRNA AC126763.1 may be a potential biomarker and therapeutic target for gastric cancer.
