| Objective To explore the effect of tumor targeting drugs Cetuximab and Gefitinib on natural killer cells(Natural killer cells,NK)tumor killing effect.In order to obtain a more effective and feasible effect of killing tumor cells: initially,to explore the effect of different concentrations of NK cells combined with different tumor-targeted drugs on the killing efficiency of colon cancer cells;next,to explore the tumor-killing efficiency of different concentrations of tumor-targeted drugs;finally,to explore the mechanism through which NK cells combined with tumor-targeted drugs destroy colon cancer cell lines.Methods Three qualified colon cancer cell lines with high expression of EGFR,Caco-2,HT-29,and Lovo,were used for cell culture through the processes of cell recovery,inoculation,medium replacement,passage,and cryopreservation.1.The growth inhibitory effect of gefitinib on three human colon cancer cell lines,Caco-2,HT-29 and Lovo,was detected by cell proliferation and cytotoxicity assay(CCK-8method).2.The growth inhibitory effect of cetuximab on Caco-2,HT-29 and Lovo three human colon cancer cell lines was detected by cell proliferation and cytotoxicity detection method(CCK-8 method).3.The growth inhibitory effect of gefitinib,cetuximab combined with NK cells on Caco-2,HT-29 and Lovo three human colon cancer cell lines was detected by cell proliferation and cytotoxicity assay(CCK-8method).4.Using the cell proliferation and cytotoxicity detection method(CCK-8method)to detect the growth inhibitory effect of NK cells on Caco-2,HT-29 and Lovo three human colon cancer cell lines under different ratios of effector cells and target cells.Result 1.Gefitinib can inhibit the proliferation of three colon cancer cells,Caco-2,HT-29 and Lovo.Gefitinib has been proved to be effective in the toxicity of colon cancer cells in a specific concentration range.2.Cetuximab can inhibit the proliferation of three colon cancer cells,Caco-2,HT-29 and Lovo.With the increase of cetuximab drug concentration,the proliferation rate of each colon cancer cell decreases faster.3.Cetuximab combined with NK cells can kill three advanced colon cancer cells,Caco-2,HT-29,and Lovo,and the combination of cetuximab concentration is 200ug/ml-300ug/ml NK cells can kill colon malignant tumor cells more effectively.4.Gefitinib combined with NK cells can kill three advanced colon cancer cells,Caco-2,HT-29,and Lovo,and the combination of gefitinib concentration is0.5ug/ml-2.5ug/ml NK cells can kill colon malignant tumor cells more effectively.Conclusion NK cells have a strong lethal activity against colon malignant tumor cells,and gefitinib can enhance this lethal effect,which may have certain therapeutic prospects in advanced colon cancer.Increasing the concentration of cetuximab will significantly improve the assisting effect of NK cells in killing colon cancer cell lines,which may be related to the drug resistance mechanism. |
