Myocardial ischemia(MI)is a cardiovascular disease in which the heart is deprived of blood and oxygen,leading to problems with the energy metabolism of the heart muscle,untreated and unprevented diseases such as angina pectoris and irregular heart rate can occur,and long-term MI can lead to irreversible damage to the heart such as myocardial infarction.The search for development of dual-use resources with anti-MI effects from Chinese medicine is of great practical importance.Acanthopanax sessiliflorus(Rupr.et Maxim.)Seem.is a medicinal plant of the genus Acanthopanax in the Araliaceae family,is a high-quality medicinal and food resource in the Changbai Mountain.The Acanthopanax sessiliflorus leaves are the medicinal part of the Acanthopanax sessiliflorus,which contains many chemical components and a high nutritional value,it is one of the optional raw materials for "treating diseases",it has been shown to have cardioprotective effects such as anti-anginal,anti-thrombotic,anti-platelet agglutination,hypolipidemic and prevention of atherosclerosis in rats,potential for the development of anti-MI adjuvant therapy products with promising applications.In this thesis,a study on the cardioprotective effect of Acanthopanax sessiliflorus leaves extract on isoproterenol(ISO)induced acute MI in rats was carried out,combined with untargeted metabolomics techniques,endogenous differential metabolites were screened and identified,metabolic pathways were analysed and their anti-MI mechanism action was initially explored.At the same time,development of an instant tea based on Acanthopanax sessiliflorus leaves to increase the added value of Acanthopanax sessiliflorus leaves products and expand medicinal and food uses of Acanthopanax sessiliflorus leaves.The main research elements and results are as follows:(1)Study on the anti-myocardial ischemic effect of Acanthopanax sessiliflorus leavesPreparation of two Acanthopanax sessiliflorus leaves extracts(aqueous extract and alcoholic extract),to examine the effects on the electrocardiogram,myocardial triple enzymes,oxidative stress indicators and histomorphology changes in rats with ISO-induced MI.The results of pharmacological experiments showed that the Acanthopanax sessiliflorus leaves extract had good anti-MI effects.Compared to the model group,prophylactic intervention with Acanthopanax sessiliflorus leaves extract significantly slowed heart rate and reduced ECG ST-segment fluctuations;significantly inhibited lactate dehydrogenase(LDH),creatine kinase(CK),aspartate aminotransferase(AST),malondialdehyde(MDA)and cardia troponin T(c Tn T)levels,significantly increased superoxide dismutase(SOD)and catalase(CAT)activities,and significantly improved myocardial tissue morphology abnormalities.The effect of the alcoholic extract of Acanthopanax sessiliflorus leaves was better than aqueous extract.(2)Analysis of the anti-myocardial ischemic effect of Acanthopanax sessiliflorus leaves based on untargeted metabolomicsA UPLC-Orbitrap-MS combined with untargeted metabolomics technique was used to screen the serum and heart samples of rats before and after the intervention with Acanthopanax sessiliflorus leaves,identify the metabolic pathways of the differential metabolite intervention and to explore the potential mechanism of action of Acanthopanax sessiliflorus leaves for its anti-MI activity.The results of untargeted metabolomics studies showed that Acanthopanax sessiliflorus leaves significantly modulated the abnormal alterations of 13 endogenous metabolites caused by MI,the differential metabolites affect a total of 16 metabolic pathways,the linoleic acid metabolic,taurine and hypotaurine metabolic and arachidonic acid metabolic were the three highest weighted metabolic pathways,which were judged to be possibly related to the anti-MI mechanism of action of Acanthopanax sessiliflorus leaves.(3)Optimization of the preparation process and quality evaluation of Acanthopanax sessiliflorus leaves instant teaTo determine the best preparation process of Acanthopanax sessiliflorus leaves instant tea using single factor and response surface methodology,and evaluated by sensory evaluation,quick solubility,the quality of the instant tea was evaluated by sensory evaluation,quick solubility,stability determination and active ingredient content determination.The optimum preparation process of instant tea from Acanthopanax sessiliflorus leaves is: crushing degree 80 mesh,ethanol concentration 60%,material to liquid ratio 1:30 g/m L,extraction time 45 min,extraction temperature 55°C,extraction 2 times,the yield is 36.25%.The highest sensory evaluation score(93.8 points)was obtained for the instant tea of prepared under the shade drying method,the stability and instant solubility met the standard,and the contents of total flavonoids,total Chiisanoside and tea-polyphenols were 98.46 mg/g,283.19 mg/g and 15.60%.In summary,this paper conducted a study on the anti-MI effect and mechanism of Acanthopanax sessiliflorus leaves,and developed an instant tea with good sensory evaluation,quick solubility,stability standards and clear content of active ingredients,which provides strong data support for the further development and utilization of Acanthopanax sessiliflorus leaves resources. |
