A large number of microorganisms are introduced into the growing environment and processing process of traditional Chinese medicinal materials.The growth and reproduction of microorganisms in medicinal materials can have many adverse effects.On the one hand,microbial contamination can reduce the effectiveness of medicinal materials,and on the other hand,microorganisms and their metabolites can also harm human health.As an important part of the production of traditional Chinese medicinal materials,it is necessary to consider the particularity of medicinal materials when choosing sterilization methods,which not only effectively kill microorganisms but also ensure the safety,effectiveness and stability of medicinal materials.There are many sterilization techniques for medicinal materials,but they all have different degrees of problems,so it is necessary to study the sterilization techniques suitable for the characteristics of traditional Chinese medicine.High Temperature Short Time(HTST)is a new type of traditional Chinese medicine materials sterilization technology.which has the advantages of short time consumption,lower energy consumption,health and safety and environmental friendly compared with tradition sterilization technology.At present,this technology is mostly used in the food industry,and its health and safety of food has been recognized.However,there are few studies on the application of HIST in the sterilization of traditional Chinese medicine materials.In this paper,taking Astragalus Radix and Cortex moutan as the research object,After the initial processing of the original medicinal materials,based on the CRITIC method,the process parameters of HTST of Astragalus Radix and Cortex moutan the effect on the quality of Astragalus Radix and Cortex moutan were inspected from multiple dimensions.Orthogonal design was used to optimize the sterilization process parameters of medicinal materials.The sterilization temperature,sterilization time and the particle size of the medicinal materials were inspected,and the sterilization rate,compound content and free radical scavenging rate of 1,1-diphenyl-2-picrohydrazine(DPPH)were determinated.The effects of sterilization on the three indexes were evaluated by intuitive and variance analysis.The samples before and after sterilization were inspected by multi-index content test,antioxidant activity and fingerprint.Antioxidant activity was determined by DPPH free radical scavenging ability,total reducing power of potassium ferricyanide and free radical scavenging ability of2-(4-Carboxyphenyl)-4,4,5,5-Tetramethyl-Imidazoline-l-Oxide-3-Oxidepotassium(PTIO).The relationship between common peaks and antioxidant activity was analyzed by partial least squares regression analysis.The results showed that the sterilization temperature was a significant factor(P<0.05).The optimal sterilization process parameters of Astragalus Radix were determined as: sterilization temperature170±2℃,sterilization time 5s,pulverized particle size not more than 24 mesh.The optimal sterilization process parameters of Cortex moutan were determined as:sterilization temperature 160±2℃,sterilization time 5s,pulverized particle size not more than 24 mesh.The microbial standard test of the three batches of samples after sterilization according to the optimized process all met the requirements of Pharmacopoeia of the People’s Republic of China.The contents of compound contents and DPPH· free radical clearance rate had no significant change compared with that before sterilization.After sterilization,the fingerprint similarity of the samples was more than 0.9.The fingerprint was correlated with DPPH free radical scavenging ability to carry out the spectral dynamic study,and the quality evaluation system was improved.The HTST process has short sterilization time and high efficiency,which is suitable for the sterilization of Astragalus Radix and Cortex moutan.The effects of HTST on the quality of Astragalus Radix and Cortex moutan were systematically evaluated by multi-dimensional indexes including measurement,fingerprint and antioxidant activity,which provided experimental basis for the application of HTST technology in the production of Astragalus Radix and Cortex moutan.
