Research Objectives: With the introduction of the 2021-2025 national fitness plan,more people are engaged in daily fitness activities,and exercise fatigue is also accompanied by it,and how to alleviate exercise fatigue has become a hot spot in current research.As an annual herb,Ghost Needle has the natural advantages of wide resources and low toxicity,and has been studied a lot.Existing studies have shown that Ghost Needle has the effects of protecting the liver,kidney,lowering blood sugar,etc.,and has the effect of alleviating oxidative stress.Therefore,this study attempts to reveal the role of the Chinese herbal herb Ghost Needle in anti-exercise fatigue.Research methods: In this study,60 mice were randomly divided into blank group,exercise exhaustion group,control group(VC:40mg/kg)and Needleweed alcohol extract low-dose,medium-dose,high-dose(40,80,160mg/kg/ kg)groups for 26 days of gavage intervention.Open field tests(upright in the middle,upright in the periphery,lying on the arm,grooming,fecal count,total distance explored in the central area,total distance explored in the surrounding area)were used to investigate whether the ethanol extract of Stipposito probiformis caused anxiety in mice.The anti-fatigue effects of ethanol extract of Stipa officinalis were determined by exhaustive swimming with weight and fatigue rotating rod test.The elevated cross maze and tail suspension tests were used to explore whether exhaustive swimming with weight and rotating rod with fatigue brought anxiety and depression to mice.Finally,the fatigue model was prepared by exhaustive swimming with weight.The contents of lactic acid(LA),urea nitrogen(BUN),glucocorticoid(GC)and creatinine(Cr)in serum and the activities of lactate dehydrogenase(LDH)and creatine kinase(CK)in liver and muscle were determined.The contents of liver/muscle glycogen(HG/MG),malondialdehyde(MDA),oxidized glutathione(GSSG)and reduced glutathione(GSH)were determined)content and activities of superoxide dismutase(SOD)and reduced glutathione(GR)were detected,so as to explain the mechanism of anti-fatigue effect of ethanol extract of Stipa bibium from the aspects of energy material reserve,metabolite accumulation and oxidative stress relief.Results of the study: 1.The ethanol extract of Stipa phobium has no effect on the growth and development of mice.There was no significant difference in body weight among the6 groups(P>0.05).2.The ethanol extract of Stipa phobium did not affect the anxiety of mice.The open field test results showed no significant difference among the 6groups of mice(P>0.05).3.The ethanol extract of Stipa phobium can prolong the exhaustive swimming time of mice,showing certain anti-fatigue effect.Compared with the exhaustive exercise group,the ethanol extract of Stipa potentilla could significantly prolong the exhaustive swimming time of mice(P<0.01).4.The experiment of exhaustive swimming with heavy weight and rotating rod with fatigue did not bring depression and anxiety to mice.There were no significant differences in the results of tail suspension test and elevated cross maze test among the 6 groups of mice(P>0.05).5.The ethanol extract of Stipa bivata can delay the accumulation of metabolic waste in the body of tired mice,improve glycogen reserve,and alleviate oxidative stress damage of muscle and liver.Compared with the exhaustive exercise group,the ethanol extract of P.hoppensiflora could decrease the contents of LA(P<0.01),BUN(P<0.05)and Cr(P<0.05)in blood of tired mice,increase the content of HG in muscle(P<0.05),and decrease the content of MDA in muscle and liver(P<0.05).The ratio of GSH/GSSG in liver and muscle was increased(P<0.05).Conclusions: In this study,the exercise fatigue model of mice was prepared by exhaustive swimming with weight-bearing force,and the anti-acute fatigue effect of ethanol extract of Stipa bivata was studied,and the following conclusions were drawn:1.The ethanol extract of Stipa phobium did not affect the normal growth of mice.2.The ethanol extract of Stipa phobium can significantly prolong the exhaustive swimming time of mice.3.The alcohol extract of Stipa biophyllum can significantly reduce the levels of blood metabolites BUN,LA and Cr in exhausted swimming mice;4.The alcohol extract of Stipa biophyllum can significantly increase the content of HG in muscle tissue;5.The ethanol extract of Stipa biophyllum can significantly reduce MDA content in muscle and liver tissue,and increase the ratio of GSH/GSSG.In conclusion,the ethanol extract of Stipa phobium has obvious anti-fatigue effect.This may be related to reducing the accumulation of metabolites,increasing glycogen reserves and enhancing the antioxidant capacity of the body.
