| Objective: To evaluate the quality of two kinds of Ganoderma lucidum spores powder from the aspects of appearance,identification,routine inspection items,breaking rate,inorganic element content and main component content.The extraction process of polysaccharides from Ganoderma lucidum sporangium powder was optimized to obtain the best extraction process,and the monosaccharide composition of the two polysaccharides was determined.The liver injury model of paracetamol was established and the hepatoprotective effect of sporangium polysaccharide was evaluated.Methods: The quality of ganoderma lucidum spore powder was evaluated by referring to the method under Ganoderma lucidum in 2020 edition of Chinese Pharmacopoeia,and the wall breaking rate and other inorganic elements of two Ganoderma lucidum spore powder were determined.The extraction process of polysaccharides from Ganoderma lucidum sporangium powder was optimized by response surface method,and the monosaccharides in polysaccharides from two kinds of sporangium powder were determined by PMP pre-column derivations combined with high performance liquid chromatography.After 14 days of preadministration of sporangium powder polysaccharide,the protective effects of two ganoderma lucidum sporangium polysaccharide on liver injury of APAP were investigated using paracetaminophen as the model drug.Results: 1.The spores of Ganoderma lucidum in the two substrates were brown and loose when the walls were not broken.After breaking the wall,the color is deepened,the texture is moist,and there is grease taste.In the chromatogram of the test product,fluorescence spots of the same color were shown at corresponding positions with the chromatogram of the control medicinal materials.The water content of ganoderma spores powder in wood substrate and substitute substrate were 8.69% and 6.31%,the total ash content was 0.87% and 0.78%,the acid insoluble ash content was 0.32% and 0.29%,the heavy metal elements As,Hg and Cd were not detected,there was no significant difference between Cr and Pb elements,but there was some difference between Cu elements.The substitute matrix was significantly higher than that of wood substrate(p<0.05).Other inorganic elements,Ba and Sn contents were significantly different(p<0.05).Ba in wood substrate sporangium powder was significantly higher than that in substitute substrate sporangium powder,while Sn was significantly lower than that in substitute substrate sporangium powder.In addition,Mg and Na in the substrate sporangium powder were significantly higher than those in the wood substrate sporangium powder(p<0.01),but there was no significant difference in other elements.The extracts were 7.25% and 6.92%,respectively.Polysaccharide content was 1.334% and 1.221%,and total triterpene content was3.60% and 4.02%,respectively.2.In the extraction process optimization experiment of Ganoderma lucidum sporangium polysaccharide,the single factor experiment results were as follows: solid-liquid ratio 15:1(m L:g),extraction time 1.5 h and extraction temperature 80℃.The response surface optimization experiment was carried out based on these results,and the optimal extraction rate was 1.85%.In the determination of the monosaccharide composition of Ganoderma lucidum sporangium polysaccharide,the polysaccharides of the two substrates were composed of mannose,rhamnose-rhamnose,glucose,xylose and fucose.The average monosaccharide content of GLP segment was 2.28,1.44,29.82,1.60 and 0.601(mg/100mg),respectively,and the mass ratio was 1.00:0.69: 13.07:0.84:0.29;The average content of monosaccharides in GLP substitutes were 2.62,2.87,54.33 and 1.951.15(mg/100mg),respectively,and the mass ratio was 1.00:1.20:20.72:0.89:0.48.3.Both of the two substrates of Ganoderma lucidum sporangium polysaccharide can improve the liver damage caused by APAP,and the effects are similar.Overall,the difference is not significant.At the same time,the m RNA expression of caspase3 and Bax and the inhibitory factor Bcl-xl were decreased,so as to inhibit apoptosis and protect liver.Conclusions: The quality of Ganoderma lucidum spore powder of the two substrates was different after breaking the wall.The content of water,ash,acid insoluble ash,extract and polysaccharide of Ganoderma lucidum spore powder of wood segment cultivation was higher,and the content of triterpene of ganoderma lucidum spore powder of ganoderma lucidum spore powder was higher.There were significant differences in the content of copper(P<0.05),significant differences in the content of other inorganic elements,Ba and Sn(p<0.05),and extremely significant differences in Mg and Na elements.The optimum extraction conditions of sporangium powder polysaccharides were as follows: liquid-solid ratio 17.6:1,extraction time1.8 h,temperature 85.9℃,extraction yield 1.85%.The two kinds of sporangium sporangium polysaccharides were composed of mannose,rhamnose,glucose,xylose and fucose,but the content and substance quantity ratio were different.In the evaluation of liver protective effect,two kinds of ganoderma lucidum spores powder polysaccharide can protect liver by regulating oxidative stress and apoptosis of liver cells. |
