Physalis Calyx Seu Fructus is the dried persistent calyx with sour pulp or fruit of solanaceous plants.It is a kind of dual-use food and medicine plant,which is described in Chinese Pharmacopoeia as having the effects of clearing heat and detoxifying,promoting pharynx and phlegm,diuresis and drenching.As a natural flavonoid compound,galuteolin exists in many plants and has strong antibacterial,anti-inflammatory,antipyretic and analgesic effects.It also plays a good role in anti-virus and anti-tumor.It is also the main active ingredient of Physalis Calyx Seu Fructus.Studies have shown that it has various pharmacological activities,such as anti-inflammatory,antibacterial,antioxidant,etc.,and has good application prospects.The pharmacological activity of the ethanol extract of Physalis Calyx Seu Fructus was studied by cell experiment,antibacterial and antioxidant experiment in this paper.Based on the liquid chromatography technology,the extraction process of the calyx alcohol extract was optimized using single-factor and response surface experiments,and the formulation of the calyx alcohol extract was prepared as a portable granule using the calyx alcohol extract as raw material.The experimental results were as follows.1.Study on the biological activity of Calyx lucidum alcoholic extract: Calyx lucidum alcoholic extract has good anti-inflammatory effect.It can inhibit the release of NO in inflammatory cells and effectively reduce the level of inflammatory factors TNF-α and IL-6,while the level of IL-10 is increased.And it has certain antibacterial effect.Its minimum inhibitory concentration for Staphylococcus aureus is 6.4 mg/m L and for E.coli is 12.8 mg/m L,and it can reduce the growth curve of E.coli and Staphylococcus aureus within 5 hours.The alcoholic extracts of Calyx lucidum had antioxidant ability.And its scavenging power against DPPH free radicals showed dose dependence.It has good biosafety and does not kill NIH-3T3 cells at 400 μg/m L.2.Optimization of the extraction process: The optimal extraction conditions were obtained by single-factor experiments,with a material-to-liquid ratio of 1:15,extraction time of 30 min,two times of extraction,ethanol concentration of 60%,and sonication temperature of 50 ℃.By using the Plackett-Burman method,the factors that had no significant effect on the extraction rate,i.e.,the material-liquid ratio and the number of extractions,were screened out.The results of the response surface experiments were analyzed and the optimized process conditions were: extraction time of 30 min,ethanol concentration of 63%,and sonication temperature of 50°C.The extraction rate was 0.805%.3.Preparation of mint alcohol extract granules: Through the preliminary screening of excipients and corrective agents,the final formulation of granules was optimized by using orthogonal experiments,and the final formulation was set as extract: dextrin: sucrose = 1 : 1.6 : 2.4.The mint brain addition was 0.5%,and the wetting agent was 70% ethanol.The prepared granules met the requirements of the Chinese Pharmacopoeia for granules.This study proved that the alcohol extract of Calyx adenophora had anti-inflammatory,antibacterial and antioxidant abilities,and optimized its extraction process to prepare granules which were convenient to carry and had unique flavor in line with the Chinese Pharmacopoeia.
