Objective: Lnc RNAs are usually involved in different physiological and pathological processes.It has been found that Lnc RNAs play an important role in the metastasis,proliferation,apoptosis,regulation of tumor microenvironment and promotion of drug resistance of breast cancer cells.We aimed to investigate the potential role of RP11-358L4.1 to provide a new theory for the mechanism of the development and occurrence in breast cancer.Methods: We first cultured and measured the expression of RP11-358L4.1 in four breast cancer cell lines,and then detected the proliferative activity,migration ability,cell cycle-related protein expression and sensitivity to carboplatin drugs in breast cancer cells after knocking down the expression level of RP11-358L4.1 by si RNA.Results:We found that the expression of RP11-358L4.1 was highly in BT-474 and MCF-7 breast cancer cell lines.After knocking down the expression of RP11-358L4.1 in these two cell lines,the migration ability and proliferative activity of cells were significantly inhibited compared with the control group(P<0.05).In addition,after knocking down the expression of RP11-358L4.1,levels of cell cycle-related proteins such as CDK1,CDK2,Cyclin B and CDC25 were significantly lower than those in the control group while the expression levels for P53 and P21 were elevated(P<0.05).This study found that after knocking down RP11-358L4.1,the sensitivity of the experimental group to carboplatin was significantly increased compared with the control group(P<0.05).Conclusion: RP11-358L4.1 can promote the proliferative activity and migration ability of breast cancer cells,and inhibit the sensitivity of breast cancer cells to chemotherapy drugs and affect the expression of cell cycle-related protein genes.
