| Objective: The expressions of hsa_circ_0023255 and hsa_circ_0006391 in the peripheral blood of the type 2diabetes mellitus group(T2DM group)and healthy control group(NC group)in Han nationality were compared.To analyze the correlation between hsa_circ_0023255,hsa_circ_0006391 and type 2 diabetes,and to provide ideas for the pathogenesis of type 2 diabetes.Methods: 1.According to the inclusion and exclusion criteria,20 Han patients with type 2 diabetes mellitus and 20 healthy controls were randomly selected.Basic data such as gender,age,BMI,SBP,DBP,and peripheral blood samples of all subjects were collected.2.An automatic biochemical analyzer detected the levels of FPG,2H-PG,TC,TG,LDL-C,and HDL-C.FINs and CP were detected by Roche electrochemical immunoassay analyzer.HOMA-IR and HOMA-β were calculated.The expression of hsa_circ_0023255 and hsa_circ_0006391 was detected by real-time fluorescence quantitative PCR.Bioinformatics analysis of the biological functions and enriched biological pathways of hsa_circ_0023255 and hsa_circ_0006391.Results: 1.There were no significant differences in gender,age,BMI,SBP,DBP,TG,LDL-C,and CP between the NC group and T2 DM group(P > 0.05).Compared FPG,2H-PG,TC,FINs,and HOMA-IR in the T2 DM group were higher than in the NC group L-C and HOMA-β were lower(P < 0.05).2.Compared with the NC group,the relative expression levels of hsa_circ_0023255 and hsa_circ_0006391 in the T2 DM group were higher than those in the NC group(P < 0.05).3.Correlation analysis showed that the relative expression levels of hsa_circ_0023255 and hsa_circ_0006391 were correlated with FPG,2H-PG,TC,FINs,and HOMA-IR,and HOMA-β(P < 0.05).4.Logistic regression analysis showed that the increased relative expression levels of hsa_circ_0023255 and hsa_circ_0006391 were risk factors for type 2 diabetes in the Han population.5.ROC curve showed that hsa_circ_0023255 and hsa_circ_0006391 had a good diagnostic value for type 2 diabetes in the Han population.6.GO functional analysis found that hsa_circ_0023255 and hsa_circ_0006391 may be involved in biological processes such as nuclear transcription or cell cycle.KEGG pathway analysis showed that hsa_circ_0023255 and hsa_circ_0006391 may be involved in the PI3K-Akt signaling pathway.Conclusion : 1.Compared with the healthy group,the expression levels of hsa_circ_0023255 and hsa_circ_0006391 were increased in the Han nationality patients with type 2 diabetes.2.hsa_circ_0023255and hsa_circ_0006391 have potential clinical diagnostic value for type 2 diabetes in Han population,and hsa_circ_0023255 has better clinical value for the early diagnosis of type 2 diabetes. |
