| Objective: To establish a fingerprint and content determination method for Quercus salicina Blume extract;To study the embryonic developmental toxicity and cardiac toxicity of Quercus salicina Blume extract in zebrafish;To explore the effect of Quercus salicina Blume extract on kidney stones.Methods: Based on relevant references and the methods specified in the 2018 edition of the "The Japanese standards for non-Pharmacopoeial crude drugs",the solvent extraction,ultrasonic assisted extraction,and chromatography(HPLC)were used to establish fingerprint spectra and content determination;the biological safety of Quercus salicina Blume extract was evaluated using a zebrafish embryo model;the effect of Quercus salicina Blume extract on wae studied kidney stones using rat renal tubular epithelial cells(NRK-52E)in vitro.Results: The fingerprint establishment of Quercus salicina Blume extract was completed and were identified five monomer compounds.The content determination results show that each gram of Quercus salicina Blume extract contains 0.80 mg of ellagic acid.The Quercus salicina Blume extract above 0.375 mg/m L resulted in a shortened body length of zebrafish,pericardium edema and myocardial cell apoptosis,delayed absorption of yolk sac,some phenomena such as incomplete development of fish bladder.In vitro cell experiments,the cell viability was approximately 60% after modeling with 0.08 mg/m L oxalic acid;After administration of low,medium,and high concentrations of Quercus salicina Blume extract,the oxalate crystals in its cell morphology decreased;Cell viability has increased;The levels of cytotoxicity,cell apoptosis,cellular lipid peroxidation damage,and reactive oxygen species all showed a decreasing trend.Conclusion:The fingerprint and content determination provide a guarantee for the quality contend of Quercus salicina Blume extract.The embryonic development and cardiotoxicity of the Quercus salicina Blume extract were not toxic to fish accordingly.Through in vitro cell experiments,it has been proven that Quercus salicina Blume extract has inhibitory effects on the formation,growth,and accumulation of oxalate crystals,as well as inhibitory effects on oxidative stress caused by oxalic acid. |
