Objective: To explore the preventive and therapeutic mechanism of Buyang Huanwu decoction on brain damage in type 2 diabetes mellitus,the levels of blood glucose,body weight,water intake,food intake,brain pathomorphology,expression of BDNF and ERK1/2protein and oxidative stress were observed and compared in each group,to explore the mechanism and approach of treatment.Methods:SPF C57 BL 6J mice were fed with a high fat and high sugar diet for 4 weeks to establish the type 2 diabetic brain damage model.The Streptozocin were randomly divided into 5 groups,metformin group,model group,low,middle and high dose groups of Buyang Huanwu decoction,10 rats in each group.The normal group and the model group were given the same amount of normal saline,once a day.The model group,metformin group and low,middle and high dose groups of Buyang Huanwu decoction were fed with high-fat and high-sugar diet,all patients were sacrificed after 8 weeks of treatment.The pathological changes of hippocampus were observed by HE staining,and the contents of MDA and GSH in serum were detected by ELISA kit.The expression levels of BDNF protein,ERK1/2 and p-ERK protein were detected by Western blot.Results:(1)Compared with the normal group,the weight of mice in the model group was significantly lower(P<0.05),and the water intake,food intake and blood glucose were significantly higher(P < 0.05),the weight of mice in each treatment group was significantly increased(P<0.05),and the water intake,food intake and blood glucose were significantly decreased(P < 0.05);(2)The pathological morphology of the hippocampus showed that the hippocampal neurons in the model group were pyknosis,decrease and necrosis in different degrees;(3)Compared with the normal group,the level of serum MDA in the model group was significantly increased(P < 0.01),the activity of GSH was significantly decreased(P < 0.01),and the level of MDA in the treatment groups was significantly decreased(P<0.05),GSH activity increased in western medicine group and middle dose group(P<0.05)(4)Compared with the normal group,the expression of BDNF protein and p-ERK1/2 in the brain of the model group were significantly decreased(P<0.05),and the expression of BDNF in the treatment groups were significantly increased(P<0.05).The expression of p-ERK1/2 was significantly increased in all treatment groups(P< 0.05),and the effect was most obvious in the high-dose group of Buyang Huanwu decoction.It is suggested that Buyang Huanwu decoction can activate ERK1/2 pathway,promote its phosphorylation,promote the expression of BDNF protein,reduce neuron apoptosis and improve synaptic dysfunction.Conclusion:Buyang Huanwu decoction can improve the typical symptoms of diabetes such as excessive drinking,food intake and weight loss in mice with diabetic brain injury.Lower Blood Sugar.Improve oxidative stress;activate ERK1/2 pathway,promote its phosphorylation,promote BDNF protein expression,reduce neuron apoptosis,improve synaptic dysfunction.To alleviate the brain damage caused by diabetes.Buyang Huanwu decoction has preventive and therapeutic effects on brain injury in T2 DM mice,and its mechanism may be related to activation of BDNF/ERK1/2 signaling pathway.
