Objective: 1.This study explored the mechanism of electroacupuncture in the treatment of Mild cognitive impairment caused by sleep deprivation through the effect of single point(Baihui),acupoint compatibility(Baihui,Shenmen,Sanyin-jiao)on the content of hippocampus and cognitive related neurotransmitters Ras,c-raf,ERK1/2 in sleep deprivation model rats.2.To provide a new idea for clinical acupuncture and moxibustion treatment of mild cognitive dysfunction caused by insomnia by comparing the dose effect difference of electroacupuncture single point and acupoint combination intervention on cognitive function of rats after sleep deprivation.Methods:1.Forty healthy male SD rats were randomly divided into 4 groups with 10 in each group.They were blank group,model group,and treatment group.The treatment consists of two groups: one group is a single acupoint group with only Baihui acupoints(referred to as the "single acupoint group"),and the other group is a Baihui Shenmen Sanyin-jiao compatibility group(referred to as the "compatibility group").2.The model group and the treatment group were given intraperitoneal injection of chlorophenylalanine(PCPA)to replicate the model.3.On the second day after successful modeling,the treatment group was given electroacupuncture treatment.The single acupoint group selected Baihui acupoint,while the compatibility group selected Baihui acupoint,bilateral Shenmen acupoint,and bilateral Sanyinjiao acupoint.The electroacupuncture was performed using a continuous wave with a frequency of 20 Hz for 30 minutes each time for 7 consecutive days;The blank group and model group were grabbed and fixed under the same conditions without any treatment.4.At the same time,two Ethology experiments were used to observe the changes of cognitive functions such as learning and memory in sleep deprivation(SD)rats before and after electroacupuncture intervention.Morris water maze includes positioning navigation experiment and space exploration experiment.The content of new Object detection(NOR)experiment includes the number,time and distance of rats’ exploration of new and old objects to detect the cognition of rats.5.The rats in each group were killed and sampled one hour after the end of the last treatment,and the cognitive related proteins in the Hippocampus of the rats were detected by Western blotting,and their expression changes were analyzed.Results:1.There were no statistically significant differences(p>0.05)in the escape latency and number of crossing platforms between each group before Morris water maze experiment training,as well as in the exploration time and resolution ratio of new objects before NOR experiment training.Two behavioral experiments were started for training.(1)Morris Water Maze The positioning navigation experiment showed that the escape latency of the model group was longer than that of the blank group(p<0.01),and the latency of the single acupoint group and the compatibility group was significantly shorter than that of the model group(p<0.05),and the latency of the compatibility group was shorter than that of the single acupoint group(p<0.05).The space exploration experiment showed that compared with the blank group,the number of times the model group rats crossed the platform was significantly reduced(p<0.01),while the number of times the single acupoint group and compatibility group crossed the platform was significantly increased(p<0.05)compared to the model group,and the number of times the compatibility group crossed the platform was significantly higher than the single acupoint group(p<0.05).(2)The NOR experiment showed that compared with the blank group,the model group rats had significantly reduced time to explore new objects and significantly reduced resolution to new objects(p<0.05);Compared with the model group rats,the treatment group rats had longer exploration time and higher resolution of new objects,and the compatibility group had longer exploration time and higher resolution than the single point group(p<0.05).2.The results of Western blot showed that compared with the blank group,the expression levels of Ras,c-raf,and ERK1/2 proteins in the hippocampal tissue of the model group decreased.Compared with the model group,the expression levels of Ras,c-raf,and ERK1/2 proteins in the hippocampal tissue of the single acupoint group and the compatibility group increased,and the compatibility group was significantly higher than the single acupoint group,indicating that the compatibility group was superior to the single acupoint group in improving the learning and memory abilities of sleep deprived rats.Conclusion:1.Electroacupuncture has a good improvement effect on cognitive impairment caused by sleep deprivation in rats,and the improvement effect of acupoint compatibility is better than that of single acupoints.2.Electroacupuncture may activate the Ras/Raf/MEK/ERK signaling pathway related to cognitive function in hippocampal neurons,and have varying degrees of impact on the protein expression of this pathway.Therefore,it is proposed that electroacupuncture may improve cognitive function by altering the neurophysiological activity of the hippocampus,promoting the differentiation,regeneration,and survival of hippocampal neurons.
