| Objective:With the acceleration of people’s life rhythm and the increase of mental pressure,the incidence rate of depression is increasing year by year.The idea of developing new antidepressants from traditional Chinese medicine compounds has attracted increasing attention due to the unique advantages of traditional Chinese medicine compounds,such as small side effects and being suitable for long-term use.Shenyi Anshen Tablet is a compound preparation designed by the research group based on the theory of traditional Chinese medicine,consisting of four traditional Chinese medicines:Yizhiren,Ginseng,Schisandra chinensis,and Ziziphus jujube kernel.This study was guided by the theory of Chinese medicine and adopted the research method of spectrum-effect relationship of Chinese medicine to investigate the antidepressant effect of Shenyi Anshen Tablet,providing a basis for its clinical application.Methods:1.Network pharmacology research:screen the gene targets and related pathways of Shenyi Anshen Tablet in treating depression through network pharmacology research,and explore the therapeutic effect of Shenyi Anshen Tablet in treating depression.2.Study on antidepressant pharmacodynamics:The model of mice was established by chronic unpredictable mild stimulation for 28 consecutive days.Through behavioral tests such as body mass,sugar-water preference test,tail suspension test,water maze test,and HE staining results of mouse hippocampus,it was determined whether the model was successful and whether Shenyi Anshen Tablet improved the depression of mice.3.Study on the spectrum effect relationship of antidepressant:HPLC was used to establish the fingerprint of Shenyi Anshen Tablet,and the gray correlation analysis method was used to establish the spectral effect relationship analysis method for the influence of Shenyi Anshen Tablet on the pharmacodynamic indicators of depression mice.The chemical components that play a pharmacodynamic role were analyzed.Results:1.Network pharmacology research:Based on the research of network pharmacology methods,it is revealed that the compound Shenyi Anshen Tablet have antidepressant effects,and play a role through multiple components,multiple targets,and multiple pathways,providing ideas for further research on the antidepressant effects of Shenyi Anshen Tablet.2.Study on antidepressant pharmacodynamics:After establishing the model of CUMS in mice,the results of behavioral test and HE staining in the hippocampus of mice confirmed that Shenyi Anshen Tablet had good antidepressant effect.3.Study on the spectrum effect relationship of antidepressant:The chromatographic conditions were determined by HPLC,and a safe and stable fingerprint method was established.The fingerprints of different batches of Shenyi Anshen Tablet were constructed,and 6 known components and 15 common peaks were identified.A method for analyzing the effect of Shenyi Anshen Tablet on the pharmacodynamic indexes of depression mice was established,and the relative correlation degree was obtained.The results showed that the contribution degree of each chemical component to each pharmacodynamic index was different.There were 10 chromatographic peaks with the correlation degree greater than 0.7,of which the correlation degree of peak 5 ginsenoside Rb1was the largest,indicating that it played a key role in the development of antidepressant efficacy.Conclusion:In this study,the antidepressant effect of Shenyi Anshen Tablet was studied,and the fingerprint of Shenyi Anshen Tablet was established;The method of network pharmacology combined with animal experiment confirmed that Shenyi Anshen Tablet has antidepressant effect;At the same time,the analysis method of spectrum effect relationship of Shenyi Anshen Tablet on the pharmacodynamic indexes of depression mice was established,which proved that many chemical components participated in the antidepressant effect of Shenyi Anshen Tablet together,providing a basis for follow-up clinical research. |
