| Objective:To investigate the effects of the invigorating the spleen,removing dampness and reducing lipid method on the general and behavioral status,monoamine neurotransmitter content,blood lipid level,vascular endothelial damage marker content and inflammatory factor level of chronic unpredictable mild stress(CUMS)rats based on lipid metabolism,Intervention with modified Suanzaoren decoction and to analyze the antidepressant effect and mechanism of the invigorating the spleen,removing dampness and reducing lipid method.Methods: SD rats were randomly divided into 6 groups,namely,blank group(Control),model group(Model),fluoxetine group(Fluoxetine),and modified Suanzaoren decoction low(SZRT-L),medium(SZRT-M),and high dose group(SZRT-H).and the depression rat model was replicated by CUMS combined with solitary feeding method,the blank group and model group were gavaged with saline,the fluoxetine group was gavaged with fluoxetine solution,and the Chinese medicine group was gavaged with modified Suanzaoren decoction with the method of invigorating the spleen,removing dampness and reducing lipid.After 21 days,the rats’ depression status was evaluated by observing the general condition,sugar and water preference and absenteeism scores;the content of monoamine neurotransmitters such as 5-HT,DA and NE,serum TG,TC,HDL-C and LDL-C,vascular damage markers such as ET-1and ICAM-1,and inflammatory factors such as TNF-α,IFN-γ,IL-1β and IL-10 were measured by ELISA.Results: 1.Compared with the blank group,CUMS rats had rough and lusterless fur,reduced number of paw grooming,reduced activity,abnormal mental status,and significantly lower scores in sugar water preference and absenteeism test(P<0.01).After treatment,compared with the model group,the rats in the low,medium and high dose groups of modified Suanzaoren decoction showed increased activity,increased number of paw grooming,soft hair,restored mental status,and significantly higher scores in sugar water preference and absenteeism test(P<0.01,P<0.05).2.Compared with the blank group,brain tissue levels of 5-HT,DA and NE were significantly decreased in CUMS rats(P < 0.01,P < 0.05),serum levels of TG,TC,LDL-C and HDL-C were significantly increased(P < 0.05,P < 0.01),and serum levels of ET-1 and ICAM-1 were significantly increased(P < 0.05,P < 0.01).01),TNF-,5-HT,DA and NE were significantly increased(P < 0.01,P < 0.05),and serum TC,LDL-C and HDL-C levels were significantly decreased in the low-dose,medium-dose,high-dose and fluoxetine groups after the intervention compared with the model group(P < 0.01,P < 0.01).Serum C and HDL-C levels were significantly decreased(P<0.05,P<0.01),serum ET-1 and ICAM-1 levels were significantly decreased(P<0.01),serum TNF-α and IFN-γ and IL-1β levels were significantly decreased(P<0.05,P<0.01),and serum IL-10 levels were significantly increased(P<0.05,P<0.01).Conclusion: 1.The Invigorating the spleen,removing dampness and reducing lipid method was able to improve the sugar-water preference and absentee field experiment scores to reduce the pleasure deficit,increase the interest in exploration and improve the depressionlike behaviour of CUMS rats.2.The Invigorating the spleen,dispelling dampness and regulating lipid method can exert antidepressant effects by increasing 5-HT,DA and NE levels in the brain.3.The mechanism of improving depression by Invigorating the spleen,dispelling dampness and regulating lipid method can reduce TG,TC,LDL-C and HDL-C levels,inhibit ET-1 and ICAM-1 vascular endothelial damage,down-regulate the levels of proinflammatory factors TNF-α,IFN-γ and IL-1β,and up-regulate the level of anti-inflammatory factor IL-10 for anti-depressant purposes. |
