Objective:To research the effect of Pi-Dan-Jian-Qing decoction on type 2 diabetes(T2DM)rat by the PTEN induced kinase 1(PINK1)/E3 ubiquitin ligase(Parkin)Signaling pathway.Methods:Forty SPF C57BL/6 rats were randomly divided into normal,model,metformin,Pi-Dan-Jian-Qing low-dose,and Pi-Dan-Jian-Qing high-dose groups,with 8 rats in each group.Except for the normal group,T2 DM rat were established by a high-carbohydrate high-fat diet and a single intraperitoneal injection of streptozotocin(STZ).The administration groups were intragastrically given the corresponding drug solution for four consecutive weeks.After the last administration,the levels of fasting blood glucose(FBG),fasting insulin(FINS),islet β cell function index(HOMA-β),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and malondialdehyde(MDA)in each group were observed and compared.Additionally,the levels of interleukin-1 β(IL-1β),tumor necrosis factor α(TNF-α),interleukin-6(IL-6),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),triglyceride(TG),and low-density lipoprotein cholesterol(LDL-C)in T2 DM rat were also observed.Finally,the protein expression of PINK1,Parkin,P62,microtubule-associated protein 1 light chain 3(LC3),cytochrome C oxidase(COXIV),voltage-dependent anionic channel protein 1(VDAC1),and mitochondrial outer membrane transporter 20(TOM20)were compared in pancreatic tissues of each group.Results:Compared to the normal group,the model group showed increased levels of FBG,HOMA-β,TC,TG,LDL-C,MDA,IL-1β,IL-6,TNF-α,COXIV,P62,VDAC1,TOM20,while HOMA-β,HDL-C,SOD,GSH-Px,PINK1,Parkin,and LC3II/I decreased significantly(P < 0.05 or P < 0.01).In comparison,the Pi-Dan-Jian-Qing decoction group showed a decrease in FBG,TC,TG,LDL-C,MDA,IL-1β,IL-6,TNF-α,COXIV,P62,VDAC1,TOM20,HOMA-β,HDL-C,SOD,GSH-Px,while PINK1,Parkin,and LC3II/I increased(P < 0.05 or P < 0.01)compared to the model group.HE staining indicated that treatment with Pi-Dan-Jian-Qing decoction improved the intensity of pancreatic acinar staining,and islet cells became clear and regular,with reduced vacuoles.Conclusions:Pi-Dan-Jian-Qing decoction can improve the function of islet β cells and inhibit their apoptosis,which may be achieved by regulating PINK1/Parkin signaling pathway.
