| Stroke is the most deadly neurological disease in China.Due to the complexity of stroke disease itself,there is a lack of specific drugs for stroke treatment and rehabilitation.Traditional Chinese medicine has been proved that efficacy for the treatment of Cerebral ischemic stroke.Da Qinjiao Decotion,composed of 16 herbs including Gentiana macrophylla,Chuanxiong rhizome and Angelica sinensis,was created by Liu Wansu,a famous physician of the Jin Dynasty,and is a classical and famous formula for the cure of Cerebral ischemic stroke.All showed good therapeutic effects,either alone or in combination with western drugs with Long-term clinical studies.In this study,we also established a cellular hypoxia-glucose deprivationreperfusion(OGD/R)model,and the protective effects were confirmed in PC12,N2 a and primary hippocampal neurons using the MTT method to detect the effects of Da Qinjiao Decotion on cell viability after OGD/R injury,as well as the LDH method to determine its lactate dehydrogenase release rate.However,it’s mechanism is still unclear in the treatment of ischemic stroke.Therefore,in this study,the mechanism of action of Da Qinjiao Decotion for ischemic stroke was investigated using a network pharmacology approach.All compounds were obtained from the TCMSP and BATMAN-TCM databases,and the active compounds were selected based on their oral bioavailability and drug-like indices.The potential proteins of Da Qinjiao Decotion were calibrated through the Uniprot database,while the potential genes for ischemic stroke were obtained from the Gene Cards database.Potential pathways associated with genes were identified by gene ontology(GO)and pathway enrichment analysis.A compound-disease-target pathway network was constructed.Subsequently,the potential mechanism of action of Da Qinjiao Decotion on ischemic stroke predicted by network pharmacological analysis was experimentally tested in the PC12 cell OGD/R model.Western blot results showed that compared with the OGD/R model,the Da Qinjiao Decotiontreated cells may be involved in regulating the MAPK signaling pathway,which in turn plays a protective role.In summary,network pharmacology prediction may be a molecular mechanistic tool when combined with experimental validation for the study of the relationship between Cerebral ischemic stroke and Da Qinjiao Decotion. |
