Study On The Mechanism Of Modified Danggui Shaoyao San On SLE With CD Mice Based On PI3K/Akt Signaling Pathway

Objective:To investigate the effect of modified Danggui Shaoyao San on cognitive impairment in mice with systemic lupus erythematosus(SLE).The levels of serum IL-6,NF-κb and the expression of PI3 K,Akt m RNA and PI3 K,Akt,p-Akt protein were measured by behavioral experiment,to analyze the possible mechanism of PI3K/Akt signaling pathway in the treatment of SLE with CD with modified Danggui Shaoyao San,and to provide reference for the treatment of traditional Chinese medicine.Method:Female MRL/lpr mice and control C57BL/6 mice were used.MRL/lpr mice were divided into model group,prednisone acetate group,modified Danggui Shaoyao San group and modified Danggui Shaoyao San + LY294002 inhibitor group,after adaptive feeding for 1 week,the animals were given the drug once a day for 6 weeks starting at 8weeks of age.Morris water maze and Y maze tests were performed before and after administration to evaluate their learning and cognitive function.The levels of IL-6 and NF-κb in serum were detected by ELISA,and the expression of genes and proteins related to PI3K/Akt signaling pathway were detected by q PCR and Western blot.Results:1.The results of Morris water maze showed that there was no significant difference in the escape latency and the times of crossing the platform between the groups(P > 0.05),but there was no significant difference in the escape latency and the times of crossing the platform between the groups(P > 0.05),compared with the model group,the escape latency of mice in the model group was significantly prolonged and the times of crossing the platform were decreased(P < 0.01),the escape latency of mice in the western medicine group,the Chinese medicine group and the inhibitor group was shortened,and the times of crossing the platform were increased(P < 0.05),and the escape latency of mice in the inhibitor group was shortened compared with that in the Chinese medicine group,the number of traversing platform increased(P > 0.05).The results of Y-maze showed that there was no significant difference in the rate of spontaneous alternation between the groups before onset and after onset(P> 0.05),but after intervention,there was no significant difference in the rate of spontaneous alternation between the groups(P > 0.05),compared with the model group,the spontaneous alternative response rate in the western medicine group,the Chinese medicine group and the inhibitor group increased significantly(P < 0.05),and the spontaneous alternative response rate in the western medicine group,the Chinese medicine group and the inhibitor group decreased significantly(P < 0.01),the rate of spontaneous alternation in the inhibitor group showed an increasing trend(P > 0.05).2.The levels of IL-6 and NF-κb in the model group were significantly higher than those in the control group(P < 0.01),the levels of IL-6 and NF-κb in western medicine group,Chinese Medicine Group and inhibitor group were significantly lower than those in Chinese medicine group(P < 0.01),and the levels of IL-6 and NF-κb in inhibitor group were significantly lower than those in Chinese medicine group(P < 0.05).3.The expression of PI3 K,Akt m RNA and PI3 K,Akt,p-Akt protein in the hippocampus of the model group were significantly higher than those of the control group(P < 0.01),the above indexes in the western medicine group,the Chinese medicine group and the inhibitor group were significantly decreased(P < 0.01),and the inhibitor group was still lower than the Chinese medicine group(P < 0.05).Conclusions:1.Modified Danggui Shaoyao San can improve the learning and working memory ability of SLE mice with CD.2.Modified Danggui Shaoyao San can decrease the levels of IL-6 and NF-κb in serum,which suggests that modified Danggui Shaoyao San can improve the state of SLE with CD mice from the level of inflammation.3.Modified Danggui Shaoyao San could decrease the transcription of PI3 K and AKT m RNA and the expression of PI3 K,Akt and p-Akt protein in mice with SLE and CD,the results suggested that the improvement of cognitive function in CD mice with SLE was related to PI3K/Akt signaling pathway,the therapeutic effect may be achieved by inhibiting the transcription of PI3 K and AKT m RNA and the expression levels of PI3 K,Akt and p-Akt proteins.
