Objective: To investigate the construction of a mouse skin aging model caused by chronic high sugar intake,and the antioxidant effect of Puerarin on aging mice.Method: Three-month-old male Kunming mice were fed normal feed and fructose(25%g/vol)drinking water to prepare a mouse skin aging model.After 56 days of continuous modeling with daily normal feed and fructose(25% g/vol)drinking water,the experimental animals were divided into a normal group,a model group,a metformin group(50mg/kg),a low-dose puerarin group(60mg/kg),and a high-dose puerarin group(120mg/kg),with 10 mice in each group except for 12 in the model group,for a total of 52 mice.Subsequently,after 28 days of continuous administration to the treatment groups,HE staining and Masson staining were performed to observe changes in mouse skin tissue and structure.Enzymelinked immunosorbent assay was used to detect the content of hydroxyproline(HYP),hyaluronic acid(HA),superoxide dismutase(SOD),malondialdehyde(MDA),and collagen IV in the skin and serum samples of each group of mice.Western blot method was used to detect the protein expression levels of collagen I,aquaporin 3(AQP3),keratin 19(K19),matrix metalloproteinase 9(MMP9),tumor necrosis factor-alpha(TNF-α),and interleukin-6(IL-6)in the skin tissue.Results: The HE staining results showed no significant inflammatory infiltration in the skin tissues of both the model group and the treatment group mice.The Masson staining results showed that compared with the normal group,the skin tissues of the model group mice exhibited significant fibrosis,which was significantly improved after treatment with puerarin.The enzyme-linked immunosorbent assay results showed that the levels of HYP,HA,collagen IV,and SOD in the skin tissues and serum of the model group mice were significantly lower than those in the normal group(P < 0.05),while the level of MDA was significantly higher than that in the normal group(P < 0.05).After low and high doses of puerarin intervention,the levels of HYP,HA,collagen IV,and SOD in the skin tissues significantly increased(P < 0.05),and the level of MDA significantly decreased(P < 0.05).In addition,after treatment with low and high doses of puerarin,the protein expression levels of collagen I,AQP3,and K19 in the mouse skin tissues were significantly increased,with statistical significance(P < 0.05).However,the protein expression levels of MMP9,TNF-α,and IL-6 in the mouse skin tissues were significantly decreased after treatment with low and high doses of puerarin,with statistical significance(P < 0.05).Conclusion: Puerarin could modulate oxidative stress damage induced by chronic high sugar intake,upregulate the expression of AQP3 and K19 in skin tissues,and subsequently increase skin collagen I and IV content to alleviate the degree of skin tissue fibrosis caused by high sugar diet.Secondly,puerarin reduced the protein expression levels of MMP9,TNF-α and IL-6,weakened the inflammatory response induced by high sugar environment,and thus delayed the skin aging induced by chronic high sugar intake in mice.
