| Purpose: Allergy Decoction,as a commonly used formula for the treatment of allergic rhinitis,has the function of dispelling wind and fixing surface,and is mostly used clinically for the treatment of various allergic diseases.There are few reports at home and abroad on whether Allergy Decoction can affect the cellular and humoral immune balance of the body while relieving the symptoms of allergic rhinitis.In this experiment,a mouse model of Allergic rhinitis(AR)was prepared to investigate the effect of Allergy Decoction on the local immune environment of the nasal mucosa,in order to provide more scientific basis for the clinical treatment of allergic diseases.Methods: Sixty-four 6-8 week old BALB/c mice of both sexes were purchased and randomly divided into 4 groups: normal control group,model control group,western medicine(loratadine)control group and Chinese medicine(allergy decoction)group,16 mice in each group.The mouse model of allergic rhinitis was established by using ovalbumin plus aluminium hydroxide sensitization method.After 7 days of treatment,nasal mucosa tissues were collected and stained with hematoxylin-eosin(HE)and toluidine blue to observe the histomorphological changes of nasal mucosa and the changes of mast cell number in each group of mice.bet,GATA-3 m RNA expressions were also detected by RT-PCR.Results.1.The AR-related behavioral scores of mice in each group: compared with the normal group,the behavioral scores of the model group were significantly higher,which was statistically significant(P<0.05);compared with the model group,the scores of the loratadine group and the allergic decoction group were significantly lower,which was statistically significant(P<0.05),and there was no significant difference between the groups(P>0.05).2.HE staining of the nasal mucosa of mice in each group showed that compared with the normal group,the epithelial cells of the nasal mucosa of mice in the model group were discontinuous,the cilia were disorganized or even detached,the blood vessels were dilated,more eosinophils appeared,and the submucosal glands were hyperplastic and the tissue edema was obvious.Compared with the model group,the inflammatory response in the loratadine and allergy decoction groups was less severe.The expression of IFN-γ and T-bet in the nasal mucosa was significantly lower(P<0.01)and the expression of IL-4 and GATA-3 was significantly higher(P<0.01,P<0.05)in the AR mouse model compared with the normal group.The expression of IFN-γ and T-bet increased significantly,and the expression of GATA-3 decreased significantly,while the expression of IL-4 tended to increase,but there was no statistical difference between the allergy decoction groups(P > 0.05).4.RT-PCR analysis of gene expression in mice nasal mucosa tissues: compared with the normal group,the expression of IFN-γ and T-betm RNA in the model group was decreased(P<0.01),and the expression of IL-4m RNA and GATA-3 was increased(P<0.01).There was a significant trend of elevation(P < 0.05),which was consistent with the Western blot results;GATA-3 expression had a significant decrease(P < 0.01),but IL-4 expression had a trend of elevation(P < 0.01).Conclusion:1.Allergy decoction can significantly reduce the local symptoms of nasal cavity and decrease the frequency of scratching and sneezing in AR mice.2.Allergy decoction can reduce the vasodilation and tissue edema of nasal mucosa and decrease the infiltration of inflammatory cells such as eosinophils in AR mice.3.Allergy decoction can reduce the expression of Th2-related transcription factor GATA-3 and increase the expression of Th1-related cytokine IFN-γ and transcription factor T-bet in nasal mucosa of AR mice.The allergy decoction can improve the symptoms of AR by reducing the expression of Th2-related transcription factor GATA-3 and increasing the expression of Th1-related cytokine IFN-γ and transcription factor T-bet in the nasal mucosa of AR mice. |
