| Objective: The expression of STAT3,PIAS3 and PIAS4 in breast cancer and adjacent breast tissue were detected and the relationship were analyzed between clinic-pathologic features,molecular typing and STAT3,PIAS3,PIAS4 to explore the role of STAT3,PIAS3 and PIAS4 in the occurrence,development of breast cancer so as to provide theoretical basis for developing new therapeutic targets and searching for biomarkers for prognosis evaluation of breast cancer.Method: Paraffin specimens and clinical data of patients with primary breast cancer who were surgically resected and pathologically diagnosed from March 2021 to September 2021 were collected from the Department of Oncology in Shengjing Hospital Affiliated to China Medical University,including 65 cases of breast cancer(52 cases of invasive ductal cancer,13 cases of ductal cancer in situ)and 14 cases of adjacent breast tissue.The expressions of STAT3,PIAS3 and PIAS4 were detected by immunohistochemical staining.The relationship between those expression and tumor diameter,lymph node metastasis and molecular typing was analyzed.Results: 1.Expression of STAT3,PIAS3 and PIAS4 in three groups: The expression of STAT3,PIAS3 and PIAS4 showed a decreasing trend in breast invasive ductal cancer,ductal cancer in situ and adjacent breast tissue.The expressions of STAT3,PIAS3 and PIAS4 in invasive ductal cancer tissues were significantly higher than those in adjacent breast tissue(P < 0.0167),there was no significant difference between other groups(P > 0.0167).2.Relationship between expression of STAT3,PIAS3,PIAS4 and clinic-pathologic features:The expression of STAT3,PIAS3 and PIAS4 were significantly increased with the increase of tumor diameter and p TNM stage(P < 0.05),and STAT3 expression in the group with lymph node metastasis was significantly higher than that in the group without lymph node metastasis(P <0.05).Multivariate analysis showed that STAT3,PIAS3 and PIAS4 increased significantly in the tumor diameter > 2cm group(P < 0.05).3.Relationship between the expression of STAT3,PIAS3,PIAS4 and molecular typing of breast invasive ductal cancer: The expression of STAT3 and PIAS4 in triple negative /Her-2 positive breast cancer tissues was slightly higher than those in Luminal breast cancer tissues,while the expression of PIAS3 in triple negative /Her-2 positive breast cancer tissues was slightly lower than those in Luminal breast cancer tissues,but the differences were not significant(P > 0.05).4.Correlation of STAT3,PIAS3 and PIAS4 expression in breast cancer:The expression of STAT3 in breast cancer was positively correlated with the expression of PIAS3 and PIAS4.Conclusions:STAT3,PIAS3 and PIAS4 proteins involved in the occurrence and development of breast cancer,and may be used as biomarkers to evaluate the progression of breast cancer and potential targets for breast cancer treatment. |
