Objective: HNF4α(hepatocyte nuclear factors 4α)is a subtype of HNF,involved in the maintenance of hepatocyte phenotype,promoting of hepatocyte differentiation and regulation of lipid and cholesterol metabolism.HNF4α expression is increased in mucinous tumors,but significance of HNF4α in the differentiation of benign and malignant mucous epithelial tumors and their origin has never been addressed.Methods: The expression of HNF4α in paraffin sections of 136 patients with mucinous tumor disease(48 cases of lung mucinous tumors,29 cases of colorectal mucinous tumors,and 59 cases of ovarian mucinous tumors)were studied by immunohistochemical staining.SPSS26.0 was used to analyze the difference in the expression of HNF4α.Results:1.There was no difference in the expression of HNF4α in benign and malignant lung mucinous tumors.The analysis of clinicopathological factors suggested that the expression of HNF4α in lung mucous epithelial tumors was not related to age,sex,and tumor size.2.In lung malignant mucous epithelial tumors,HNF4α expression was increased in KRAS mutant compared with KRAS wild type.The analysis of clinicopathological factors suggested that the expression of HNF4α in lung malignant mucous epithelial tumors was only related to the mutation of KRAS,and had no obvious correlation with other factors.3.The expression of HNF4α in colorectal malignant mucous epithelial tumors was lower than that in adjacent normal tissues.The analysis of clinicopathological factors suggested that the expression of HNF4αexpression in colorectal was only related to mucinous tumors,and had no significant correlation with other factors.4.Compared with benign and borderline mucinous tumors of the ovary,HNF4α expression was increased in malignant mucinous tumors of the ovary.The analysis of clinicopathological factors suggested that the expression of HNF4α expression in ovarian mucinous tumors was only related to the benign and malignant tumors,and had no significant correlation with other factors.5.The expression of HNF4α in lung malignant mucous epithelial tumors was lower than that in ovarian and colorectal malignant mucous epithelial tumors.Conclusion: 1.HNF4α is not useful in differentiating benign and malignant pulmonary mucous epithelial tumors.2.HNF4α is associated with the mutation of driver gene KRAS,and the expression of HNF4α is increased in patients with KRAS driver mutation.3.The expression of HNF4α is decreased in malignant mucous epithelial tumors of colorectal,which has differential and suggestive significance.4.The expression of HNF4α increased in malignant mucinous tumors of ovary,but showed no difference in benign and borderline mucinous tumors.5.The expression of HNF4αdecreased in malignant mucous epithelial tumors of lung origin,but had no difference in colorectal and ovarian origin.
