Objective: There is a lack of sensitive and specific serum molecular markers for the diagnosis of ALI/ARDS.In this study,we used bioinformatics tools combined with high-throughput sequencing dataset results to find meaningful biomarkers for the diagnosis and treatment of ARDS.Methods: In this study,we obtained high-throughput sequencing datasets(GSE114132,GSE164963,GSE175580)of lung tissues from a public database platform(GEO)in a mouse ALI model due to common causative factors,applied edge R for differential analysis to identify differential genes,and used Gene Annotation(GO,Gene Ontology),Kyoto Encyclopedia of Genes and Kyoto Encyclopedia of Genes and Genomes(KEGG,Kyoto Encyclopedia of Genes and Genomes)for enrichment analysis of these differential genes,and the screened differential genes were subjected to protein network analysis using the STRING database and software to calculate the maximum neighbor connectivity(Maximum Neighborhood The top 10 genes in terms of MNC,Maximal Clique Centrality(MCC)and Degree and the genes in the module with the highest MCODE(Molecular Complex Detection)scores were screened and the intersection was taken as the core gene for the DGIdb The core set of genes was taken as the core set,and drug prediction analysis was performed through DGIdb database.Results:A total of 224 differentially expressed genes(DEGs)were identified,including173 up-regulated genes and 51 down-regulated genes;The GO analysis of DEGs showed that the biological process(BP)was mainly involved in leukocyte migration,cell chemo taxis,positive regulation of response to external stimulus and other biological processes;The analysis of molecular function(MF)showed significant enrichment in immune receptor activity,peptidase regulator activity,G protein-coupled peptide receptor activity,peptide receptor activity,cytokine receptor activity,etc.Cellular Component(CC)analysis showed that DEGs were mainly distributed in plasma membrane raft,membrane raft,membrane microdomain,receptor complex,etc.;KEGG analysis showed that DEGs were significantly enriched in TNF signaling pathway,IL-17 signaling pathway,Cytokine-cytokine receptor interaction and other pathways.Cxcl2,ITGAM,Mmp9,TLR2,CD14 and FcγRIII were identified as up-regulated genes.Drug prediction analysis identified interactions between the core genes for Infliximab,Ibudilast,Fentanyl,Alteplase,Hydrocortisone,and Morphine.Conclusions: In this study,nine key genes of IL-6,IL-1β,ITGAM,Mmp9,Cxcl1,Tlr2,Cd14,Cxcl2,FcγRIII and one lnc RNA B130024G19 Rik were obtained by bioinformatics,among which IL-6,IL-1β,Mmp9,Cxcl1,Tlr2,Cd14 Cxcl2,and FcγRIII have now been shown to be closely associated with acute lung injury,and clinical progress has been made in intervening ARDS against their targets,while clinical studies of B130024G19 Rik and ITGAM in acute lung injury models are rare or have not been reported,and have the potential to become new markers for the treatment and early diagnosis of ALI.
