Expression And Correlation Of PIK3CA And PTEN In Sinonasal Squamous Cell Carcinoma | Posted on:2024-08-22 | Degree:Master | Type:Thesis | Country:China | Candidate:H Q Zhao | Full Text:PDF | GTID:2544307088980249 | Subject:Otorhinolaryngology | Abstract/Summary: | PDF Full Text Request | Objective: To analyze the mRNA expression of PIK3 CA and PTEN in head and neck squamous cell carcinoma tissues and adjacent tissues,and to explore the protein expression of PIK3 CA and PTEN in sinonasal squamous cell carcinoma,the relationship between PIK3 CA and PTEN and the clinicopathological features and prognosis of the tumor,and the correlation between them.In order to find reliable indicators for early diagnosis and treatment,staging,prognosis evaluation and precision medical treatment of sinonasal squamous cell carcinomaMethod:(1)The RNAseq dataset HNSC from TCGA was retrieved and downloaded through UCSC Xena(https: // xenabrowser.net/).The data sets were analyzed using R(version 4.2.1),the ggplot2 package was used to visualize the data,and the Mann-Whitney U(Wilcoxon rank sum test)test was used for statistical analysis.(2)Forty-three cases of sinonasal squamous cell carcinoma were collected,twenty cases of normal inferior concha tissues were chose as control.The protein expressions of PIK3 CA and PTEN were detected by immunohistochemistry. SPSS26.0 was used to analyze the data,and X2test was used to compare the positive expression rates of PIK3 CA and PTEN in sinonasal squamous cell carcinoma tissues and inferior concha tissues.The relationship between PIK3 CA,PTEN and clinicopathological features was analyzed by X2test or Fisher exact test.Kaplan-Meier method was used for survival analysis.Spearman rank correlation analysis was used to analyze the correlation between them.P < 0.05 was considered statistically significant.Result: The results of biogenic analysis showed that PIK3 CA mRNA expression in head and neck squamous cell carcinoma tissues was higher than that in adjacent normal tissues(P < 0.01),while PTEN mRNA expression was lower than that in adjacent normal tissues(P < 0.05).The positive rate of PIK3 CA expression in sinonasal squamous cell carcinoma tissues(76.74%)was higher than that in normal inferior turbinate mucosal tissues(15.00%),and the positive rate of PTEN expression(13.95%)was lower than that in normal inferior concha tissues(70.00%),and the differences were statistically significant(P < 0.01);the positive rate of PIK3 CA in patients with stage III~IV was higher than that in stage I~II patients(P < 0.05),and the positive rate of PIK3 CA was higher in the tumor tissues with low differentiation than in the middle and high differentiation groups(P < 0.05),and the positive rate of PIK3 CA was higher in the tissues of sinonasal squamous cell carcinoma tissues(P < 0.05),while PIK3 CA expression was not related to age,gender,smoking,or alcohol consumption(P > 0.05);PTEN expression was not related to tumor clinical stage,PTEN expression was not associated with clinical stage,differentiation,primary site,age,gender,smoking,alcohol consumption and prognosis(P>0.05);Spearman analysis showed that there was a significant negative correlation between PIK3 CA and PTEN expression in sinonasal squamous cell carcinoma tissues(r=-0.414,P<0.01);Kaplan-Meier analysis showed that the median survival time of patients in the PIK3 CA positive expression group was lower than that in the negative expression group.survival time was lower than that of the negative expression group(P < 0.01).Conclusion: The expression of PIK3 CA is up-regulated and PTEN is down-regulated in sinonasal squamous cell carcinoma and the two are negatively correlated,and PIK3 CA is associated with clinical progress and poor prognosis of patients with sinonasal squamous cell carcinoma. | Keywords/Search Tags: | PIK3CA, PTEN, sinonasal, squamous cell carcinoma, immunohistochemistry | PDF Full Text Request | Related items |
