Objective: To investigate the therapeutic effects of bailing powder on airway inflammation,airway hyperresponsiveness and airway remodeling in mice with bronchial asthma.Methods: BALB/C mice were divided into normal control group,bronchial asthma group,budesonide treatment group,low-dose bailing powder group,medium-dose bailing powder group and high-dose bailing powder group with 10 mice in each group.Among them,budesonide treatment group was intervened by nebulization and the low,medium and high dose Bailing powder groups were all intervened by gavage.The airway resistance of mice stimulated by methacholine was measured to evaluate the airway responsiveness of mice,and the total number of cells in bronchoalveolar lavage fluid(BALF)was counted.The infiltration of inflammatory cells in bronchi and alveoli of mice in each group was observed by HE staining,goblet cell metaplasia of tracheal epithelium was observed by AB-PAS staining,collagen deposition around airway was observed by Masson staining,and the expression and distribution of α-smooth muscle actin(α-SMA)in lung tissue was observed by immunohistochemical staining.Results: The airway resistance and the total number of inflammatory cells in BALF in low-dose bailing powder group,middle-dose bailing powder group and high-dose bailing powder group were significantly lower than those in asthma group(P <0.05).There was no significant difference in airway resistance between the three intervention groups and budesonide treatment group(P>0.05),but the total number of inflammatory cells in BALF was higher than that in budesonide treatment group(P<0.05).The results of HE staining,AB-PAS staining and Masson staining showed that compared with the asthma group,the number and infiltration of inflammatory cells around the airway in budesonide treatment group,low dose bailing powder group,middle dose bailing powder group and high dose bailing powder group were less than those in asthma group,the stenosis of airway lumen was not obvious,the metaplasia level of goblet cells in airway was reduced,the secretion of mucus was reduced,and the deposition range and degree of collagen around airway were reduced.The results of immunohistochemical staining showed thatα-SMA was positive in airway smooth muscle of asthmatic mice.After intervention with budesonide,low-dose bailing powder,medium-dose bailing powder and high-dose bailing powder,the expression level of α-SMA in airway smooth muscle of mice decreased and the thickening level of airway smooth muscle decreased.Conclusion: Bailing powder can reduce airway hyperresponsiveness and improve airway inflammation and airway remodeling in bronchial asthma mice.
