| Objective: To investigate the expression levels of tridomain protein-3(TRIM3)and P53 and their correlation with clinicopathologic features in non-small cell lung cancer(NSCLC).To explore the expression of TRIM3 and P53 in NSCLC and their potential clinical value.Methods: In this study,cancer tissues of 119 patients with NSCLC who underwent surgical resection of lung tumors in the First Affiliated Hospital of China Medical University from December 2018 to December 2019 were collected and tissue samples were sent to the Department of Pathology of our hospital,and corresponding adjacent tissues were used as controls.Clinicopathologic data of all cases were collected,and each clinicopathological index was grouped according to the standard.Immunohistochemical staining of TRIM3 and P53 was performed.A scoring system was developed for the interpretation of immunohistochemical results,and the expression of the slices of all cases was evaluated.The expression of TRIM3 and P53 in cancer tissues was analyzed,and the correlation between the expression levels and various clinicopathological features was analyzed.The expression of TRIM3 in NSCLC was analyzed using GEPIA database.The Kaplan-meier Plotter database was used to analyze the effect of TRIM3 expression level on prognosis of patients with NSCLC.The statistical analysis was carried out on SPSS26.0 statistical software.Results: 1.The expression of TRIM3 in NSCLC was significantly lower than that in paracancer tissues.The positive expression rate of TRIM3 was 14.3%(17/119)in NSLCLC and 88.2%(105/119)in paracancer tissues,showing significant difference(P< 0.0 1),which was consistent with the results of GEPIA database analysis.2.The expression of TRIM3 was correlated with lymph node metastasis(P=0.005),TNM stage(P=0.027),tumor differentiation degree(P=0.045)and tumor tumor size(P=0.009).It was not related to age of diagnosis,sex,smoking or drinking,degree of tumor differentiation,pathological classification of tumor and number of lesions.3.The expression level of TRIM3 was negatively correlated with Ki-67 proliferation index(P=0.002).4.The expression level of TRIM3 was different among different histological subtypes of lung adenocarcinoma(P=0.007).5.There was no significant difference in TRIM3 expression among different subtypes of squamous cell carcinoma(P=0.290).6.Kaplan-meier Plottery database analysis showed that adenocarcinoma patients with high TRIM3 expression(HR=0.54,P=0.011)and squamous cell carcinoma(HR=0.62,P= 0.0 4 5)all patients had longer overall survival(OS),adenocarcinoma patients with high TRIM3expression(HR=0.71,P=0.29)had higher relapse-free survival(RFS),and squamous cell carcinoma patients with TRIM3 expression level had no effect on RFS(P=0.29).7.The expression level of TRIM3 was positively correlated with that of PD-L1(P=0.049).Conclusion: 1.The expression level of TRIM3 in NSCLC is significantly decreased;2.In NSCLC,the expression of TRIM3 is negatively correlated with lymph node metastasis,tumor size,TNM stage and Ki-6 7 proliferation index,and positively correlated with tumor differentiation degree,and low expression of TRIM3 is correlated with poor prognosis.These results suggest that TRIM3 may play a role in cancer inhibition in NSCLC.3.The expression level of TRIM3 is different among different subtypes,which has the significance of clinical differential diagnosis. |
