| Purpose: The role of retinoblastoma binding protein 7 in esophageal cancer remains unclear.The purpose of this study was to investigate the expression level of RBBP7 in esophageal cancer and its relationship with patient prognosis,as well as the potential functional mechanism of RBBP7 in esophageal cancer cells.Methods: Using online databases of TIMER,UALCAN and GEPIA,the expression level of RBBP7 in esophageal cancer and its relationship with the prognosis of patients with esophageal cancer were analyzed.The expression level of RBBP7 in TE1 and EC109 esophageal cancer cell lines was knocked down,and the invasion ability of cells was detected by Transwell assay,the proliferation ability of cells was detected by clonogenesis assay,and the tumor spheres formation ability was detected by pellet formation assay,and compared with the control group.After The expression of RBBP7 knockout cell lines in EC109 and TE1 cells and in the control cell lines were up-regulated.Transwell assay,clonogenesis assay and pellet formation assay were performed to verify the regulatory effect of RBBP7 on esophageal carcinoma cells.GSEA was used to analyze signaling pathways related to RBBP7 regulation in esophageal cancer.Results: 1.RBBP7 expression is generally increased in malignant tumors,and RBBP7 RNA expression is also significantly increased in esophageal cancer.High expression of RBBP7 is associated with poorer survival outcomes in patients with esophageal cancer.2.In TE1 cells down-regulated by RBBP7,the invasion and proliferation of cells increased,and the tumor spheres formation ability increased;In EC109 cell lines down-regulated by RBBP7,cell invasion,proliferation and spheres formation ability of tumor cells were decreased.3.Recovery of down-regulated RBBP7 expression in esophageal cancer cells can reverse the promoting or inhibiting effects of low levels of RBBP7 on cell invasion,cell proliferation and spheres formation ability.4.RBBP7 may be involved in regulating cell cycle,spliceosome,base excision repair and other pathways in esophageal cancer.Conclusion: The expression of RBBP7 in esophageal cancer was significantly higher than that in normal tissues,and the survival prognosis of patients with esophageal cancer with high expression of RBBP7 is poor.RBBP7 may promote and inhibit the invasion process,proliferation process and stem cell characteristics of esophageal cancer cells. |
