Objective: The difference protein between the candidiemia group and the normal group was screened by TMT technology,and significant markers were found,which were verified by ELISA technology to provide new markers for more accurate and earlier diagnosis of candidiemia.Methods: Blood culture positive serum of Candida was collected,and part of blood culture positive and normal patients with the same underlying disease,same gender and similar age were selected as the experimental group and the control group to complete the TMT experiment,and the different proteins in the candidiemia group and the normal group were screened.Bioinformatics software was used to analyze the functions of the proteins screened above and select the target proteins.The qualitative and quantitative analysis of the target protein was performed in the serum of other patients with candidiemia by ELISA,so as to find new markers for clinical diagnosis of candidiemia.Results: Serum from 3 groups of positive and normal patients with the same underlying diseases,the same gender and the same age were selected as the experimental group and the control group(a total of 6 serum samples)to complete the TMT experiment.A total of 347 proteins were identified.By ANOVA analysis of variance,proteins with p < 0.05 and difference ratio greater than 1.2 times were screened as differential proteins.A total of 81 differential proteins(56 up-regulated proteins,25 down-regulated proteins)were found.Bioinformatics technology was used to conduct GO analysis and Pathway analysis of differential proteins,and 48 metabolic pathways were found to be involved.These include apoptosis metabolic pathways,actin cytoskeleton regulatory pathways,complement and coagulation cascades,platelet activation,leukocyte migration across endothelial cells,protein digestion and absorption,NOD-like receptor signaling pathway,estrogen signaling pathway,transcription disorders in cancer,metabolism,etc.According to the metabolic pathways involved in differential proteins,combined with previous literature analysis,The CP protein involved in the pathway of iron oxidation promoting apoptosis and F-actin capping protein subunit alpha involved in the formation of candida mycelium skeleton,promoted mycelium adhesion,and caused phagocytosis of macrophages and phages were identified by ELISA.It was verified that the two proteins were up-regulated in the serum of patients with candidiemia.Conclusion: The expression levels of CP protein,which is involved in iron oxidation to promote apoptosis,and F-actin-capping protein subunit alpha,which is involved in the formation of mycelial skeleton,promotes mycelial adhesion,and causes phagocytosis of macrophages and phages,are increased in the serum of patients with candidemia.It may be a new diagnostic index for candidiemia.
