Objective: Gestational diabetes Mellitus(GDM)refers to abnormal glucose metabolism in different degrees during pregnancy,but the blood sugar does not reach the level of dominant diabetes.The increase of blood glucose during pregnancy has a close impact on the pregnancy process.In particular,vascular endothelial dysfunction of GDM pregnant women may be the potential cause of adverse maternal and infant outcomes,but its specific pathogenesis is currently unclear.In this study,high flux sequencing was performed on the placenta tissue of GDM patients,and then bioinformatics was used to analyze the differentially expressed genes in GDM,which was verified by experiments to explore the mechanism of its function.Methods: Through sequencing of placental tissue of GDM patients,and then bioinformatics analysis,the differentially expressed genes related to vascular dysfunction in GDM were found,and the sample size was further expanded to verify the expression in placental tissue.Then,human umbilical vein endothelial cells(HUVEC)were cultured in vitro and divided into control group,high glucose group and high glucose transfection group.CCK8 method was used to detect cell proliferation;cell apoptosis was detected by flow cytometry;the cell migration ability was detected by scratch method;tubular formation test was used to detect the ability of cells to form tubes.Then explore its relationship with PI3K/Akt pathway.Results: Through the transcriptome sequencing of GDM placenta tissue,a total of 213 differentially expressed genes were screened,including 120 up-regulated and 93down-regulated.After literature review,the gene PDGFRβ that related to vascular endotheial function was selected as the key gene for further analysis.It was found that the expression level of PDGFRβ m RNA and protein was significantly higher in GDM pregnant women than in normal pregnant women.In the cell high glucose model,it was verified that the m RNA expression level of PDGFRβ increased and the protein expression also increased(P<0.05).After the treatment of knock down PDGFRβ,the proliferative capacity,migration capacity of the cells were restored,the level of apoptosis and tubular ability were decreased(P<0.05),and the levels of phos-PI3K/PI3 K and phos-Akt/Akt were decreased(P<0.05).Conclusion: The expression level of PDGFRβ increased in the placenta of GDM pregnant women,and PDGFRβ was distributed on and around the blood vessels in the placenta.The expression of PDGFRβ was increased in the high-glucose HUVEC model.Knocking down of PDGFRβ can restore its proliferation and migration ability,improve the apoptosis,reduce the ability to form tubes,and reduce the activity of PI3K/Akt pathway.PDGFRβ may be involved in the process of vascular dysfunction in GDM.
