Objective: The purpose of this study was to explore the expression and prognostic features of RNA m6 A reading protein EIF3 E in gastric cancer and colorectal cancer.Methods: In this study,we got 202 gastric cancer tissues,adjacent intestinal metaplasia tissues and adjacent normal tissues,meanwhile 88 colorectal cancer tissues and adjacent normal tissues was collected.We use immunohistochemical staining to investigate the expression of EIF3 E protein in the above mentioned gastric lesion tissue and colorectal tissue.We determined the role of prognosis of EIF3 E protein expression in 147follow-up visit gastric cancer patients and 85 follow-up visit colorectal cancer patients by Kaplan-Meier survival analysis.We confirmed the prognostic impact of EIF3 E protein in follow-up visit patients with gastric cancer and colorectal cancer by using Multivariate Cox regression analysis,and then we created a clinical prognostic nomogram severally according to the level of EIF3 E protein expression,to evaluate the prognosis of new patients with gastric cancer and colorectal cancer.We also explore the possible reasons that EIF3 E can lead to a different prognosis for gastric and colorectal cancer by using immuno-related analysis and GSEA enrichment analysis.Result: The expression levels of EIF3 E protein is the lowest in gastric cancer tissues while the highest in adjacent normal tissues,in adjacent intestinal metaplasia tissues is middle(P<0.0001).But the expression levels of EIF3 E protein in adjacent normal colorectal tissues were lower than that in colorectal cancer tissues significantly(P<0.0001).Among gastric cancer patients,the expression of EIF3 E protein was related to 9 clinical risk factors of gastric cancer closely,including the depth of invasion,grosstype,TNM stage,tumor size,degree of differentiation,lymphatic/venous invasion,growth pattern,lymphode metastasis and perineural invasion.And the expression of EIF3 E protein were associated with 7 risk factors for colorectal cancer patients,including growth pattern,lymphode metastasis,degree of differentiation,grosstype,the depth of invasion,TNM stage and lymphatic/venous invasion.The expression level of EIF3 E protein was a good predictor,which can independently predict the prognosis of gastric cancer and colorectal cancer patients very well(P <0.05).In addition,gastric cancer patients those had a low EIF3 E expression had a worse prognosis while colorectal cancer patients will have a better prognosis when they have a low EIF3 E expression.We creat a clinical prognosis predicted model according to the expression level of EIF3 E protein to evaluate the prognosis of new gastric cancer and colorectal cancer patients,and this model can predict the prognosis very well.EIF3 E expression was negatively correlated with macrophages in gastric cancer and positively correlated with macrophages in colorectal cancer.EIF3 E is mainly enriched in the processes related to B cell receptor(BCR),as well as the regulatory pathways of MAPK and NF-KB related to high-affinity IgE Fc receptor(FCERI)in colorectal cancer high-expression group.However,in the gastric cancer group with high expression,EIF3 E was enriched and down-regulated in the regulatory processes related to B cell receptors and the regulatory pathways of MAPK and NF-KB related to FCERI.Conclusions: The expression level of EIF3 E protein was the lowest in gastric cancer tissues,the middle in intestinal metaplasia tissues of paracancer,the highest in paracancer normal stomach tissues,but the expression level of EIF3 E protein in normal colorectal tissues was significantly lower than that in colorectal cancer tissues.The different prognostic effects of EIF3 E on gastric cancer and colorectal cancer may be related to macrophage and BCR-MAPK-NF-KB pathway.These findings suggest that EIF3 E may be a potential prognostic biomarker for patients with gastric cancer and colorectal cancer. |
