Objective:High-level ultraviolet radiation is the main reason for environmental stress.Skin overexposure to UV can cause DNA damage and activated oxygen(ROS)accumulation,which is the cause of showing sunburn,aging,skin cancer and other manifestations.Recent studies have shown that fat droplets during stress are critical to protect cell integrity and function:they areolate potential toxic lipids and proteins,maintain energy and oxidation and balance,maintain the balance of membrane and cytoles,regulate self-self Step,provide lipid as a signal medium.Perilipin 2(Perilipin 2,Plin2)is also known as fat differentiation-related proteins,which is not only closely related to oxidation stress,but also plays a crucial part in the balance of lipid droplets and lipid outflow.C/EBPA(CCAAT/Enhancer-Binding Protein-A)is one of the six members of the alkaline brightened zipper transcription factor C/EBP family.Bone marrow cells of alveolar epithelial cells and hematopoietic systems,the differences in expressive spectrum determine their important and unique role in different tissues and organs.Among the oxytic satellite cells treated with oleic acid,CEBPA skin on the back of the mouse for pathological materials.Organizational changes,use Western blot to detect CEBPPA and PLIN2 changes in mice skin tissue,immunohistochemical detection CEBPPA and PLIN2.2.Select the primary fibroblast(HDF)and embryo fibroblasts(3T3)as experimental objects.The MTS method detects different doses of UVA radiation to change the vitality of fibroblasts,and selects appropriate light intensity to build a light damage model.PCR and Western blot were used to determine the changes of CEBPA and PLIN2,and BODIPY493/503 staining was used to detect the expression of PLIN2,the original fibroblasts of human beings,has increased significantly.UVA irradiation can significantly increase the protein level of CEBPPA,and the Bodipy 493/503 visible ultraviolet radiation can increase the aggregation of cell fat droplets.5.After irradiation of different doses of UVA,22.5J/CM2 is selected for subsequent experiments for changes in the vitality of fibroblasts in mice.6.After UVA irradiation,the level of RNA,CEBPA,and PLIN2 of mice into fibroblasts,increased significantly.UVA irradiation can significantly increase the protein level of CEBPPA,which has no effect on the overall expression level of PLIN2.Bodipy 493/503 Dye visible ultraviolet radiation increases the agglomeration of cell fat droplets.Conclusion:1.The global research on lipid droplets is unevenly distributed,and the research enthusiasm has been steadily increasing,especially the research and exploration of the relationship between oxidative stress,autophagy and inflammation outside the function of lipid storage is increasing.2.Increased expression of CEBPA and PLIN2 proteins in UVA-induced mouse skin photoinjury models.3.UVA induces increased expression of CEBPA and PLIN2 in skin fibroblasts,and correspondingly increased the aggregation of lipid droplets in cells.
