Objective: Polycystic ovary syndrome is a common endocrine and metabolic disorder in women,characterized by chronic anovulation and hyperandrogenism,the main clinical manifestations are irregular menstruation,ovarian polycystic changes,infertility,obesity,Hirsutism and/or acne,depression and anxiety,etc.ceRNA is an element that can compete for binding to RNA,and usually lnc RNA and circ RNA will compete for binding to mi RNA.The purpose of this study is to mine the GEO database,select data sets related to polycystic ovary syndrome for differential gene analysis,construct a ceRNA regulatory network,analyze the biological functions of related genes,and explore the potential pathogenesis of polycystic ovary syndrome.Methods: Datasets related to polycystic ovary syndrome were searched through the GEO database using the key words "polycystic ovary syndrome(PCOS)","polycystic ovary syndrome","tissue" and "human",and the diagnostic criteria adopted the Rotterdam Consensus Criteria,and preliminarily analyzed the expression and functional enrichment of differentially expressed genes in the polycystic ovary syndrome group and the control group.In the GSE95728 dataset,the mi RNAs of the differentially expressed lnc RNAs were predicted by the mi Rcode database to obtain the lnc RNA-mi RNA interaction pairs;the mi RNA-mRNA interaction pairs were obtained through the mi RDB,mi RTar Base and Target Scan databases;After the mRNA is intersected,the ceRNA regulatory network is established using Cytoscape software,and the protein interaction network is drawn through the String online website to predict the biological functions of key genes.Results: 1.By limiting the search conditions,a total of 16 qualified data sets were retrieved,and the differential analysis was performed after sorting out.The biological processes in the GO functional enrichment analysis of the differential gene GO function enrichment analysis of the data sets from different sample sources are mainly involved in the positive regulation of cytokines,etc.;Cell components mainly involve secretory granule membranes,etc.;molecular functions mainly involve receptor binding,etc.;KEGG pathway enrichment analysis mainly involves receptor interactions between cytokines,etc.2.The mi RNA differentially expressed in more than two data sets is hsa-mi R-455-3p;The common differentially expressed lnc RNAs in the GSE95728 and GSE65746 data sets include 21.3.A total of 13 interaction relationships were established between mi RNA and mRNA from different sample sources.4.By establishing lnc RNA-mi RNA and mi RNA-mRNA interaction pairs,a ceRNA regulatory network of 26 lnc RNA-30 mi RNA-30 mRNA was constructed,and the protein interaction network involved a total of 18 nodes and 26 edges.5.30 mRNAs were enriched and analyzed through the Metascape online website,mainly involved in the regulation of lipid catabolism,leukocyte differentiation and epithelial cell proliferation;involving The pathways are mainly the regulation of the NF-k B signaling pathway and the apoptosis signaling pathway;the top-level Gene Ontology biological processes include metabolic processes,negative regulation of biological processes,and immune system processes.Conclusion: In this study,a total of 16 data sets related to polycystic ovary syndrome were detected in the GEO database.In the differential gene GO analysis,the biological process is mainly involved in the positive regulation of cytokines,etc.;the cellular components are mainly involved in the secretion of granule membranes;KEGG analysis mainly involved cytokine-receptor interactions.The functional enrichment analysis of the constructed 26 lnc RNA-30 mi RNA-30 mRNA ceRNA regulatory network mainly involves the regulation of lipid catabolism,leukocyte differentiation and epithelial cell proliferation;the pathways involved are mainly the regulation of NF-k B signaling pathway and apoptosis signaling pathway.
