Background:Hepatocellular carcinoma(HCC)is still a major challenge to human life.In-depth study of the pathogenesis of HCC may provide a reliable target for diagnosis and treatment of HCC.The ubiquitination system is a common system for regulating proteins.The balance between ubiquitination and deubiquitination may affect the biological function of tumor cells.Deubiquitination enzymes(DUBs)remove ubiquitins from proteins to regulate their function.At present,many deubiquitination enzymes have not been understood.The function of OTUD5 has not been clearly studied in HCC,and its function needs to be further studied.Objective:This study aims to reveal the expression,clinical significance and biological function of OTUD5 in HCC,screen out downstream proteins regulated by OTUD5 through unlabeled quantitative proteomics technology,and explore the specific molecular mechanism of OTUD5 regulation of HCC,laying a scientific foundation for the search for new tumor markers in HCC.Methods:1.The expression level of OTUD5 in liver cancer tissues and cells was verified by biogenic analysis and western blotting.2.In vitro functional experiments such as CCK-8 experiment and Edu cell proliferation experiment were used to prove the regulatory effect of OTUD5 on the biological behavior of hepatoma cells.3.Unlabeled quantitative proteomics techniques,Western blot and Co-IP experiments were used to prove the molecular mechanism of OTUD5’s regulation of biological behavior of hepatoma cells.4.In vivo animal experiments were conducted to verify the regulatory effect of OTUD5 on the biological behavior of hepatoma cells.Results:1.The expression level of OTUD5 in liver cancer tissues and cells was verified by biogenic analysis and western blotting.2.In vitro functional experiments such as CCK-8 experiment and Edu cell proliferation experiment were used to prove the regulatory effect of OTUD5 on the biological behavior of hepatoma cells.3.Unlabeled quantitative proteomics techniques,Western blot and Co-IP experiments were used to prove the molecular mechanism of OTUD5’s regulation of biological behavior of hepatoma cells.4.In vivo animal experiments were conducted to verify the regulatory effect of OTUD5 on the biological behavior of hepatoma cells.Conclusions:1.OTUD5 is a new deubiquitination enzyme in liver cancer cells.2.The expression of OTUD5 is up-regulated in liver cancer tissues and liver cancer cells,and LINC00674 is a potential molecular marker for poor prognosis of liver cancer.3.Interference with LINC00674 significantly inhibited the proliferation of hepatocellular carcinoma cells,suggesting that OTUD5 was a new promoter of hepatocellular carcinoma.4.OTUD5 regulates SLC38A1 protein expression and stability through deubiquitination in liver cancer cells,promoting the development of tumors.5.Down-regulation of OTUD5 can reduce the growth of HCC in mice,and OTUD5 can be used as a potential molecular marker in vivo. |
