Objective: To investigate the effects of sleeve gastrectomy on the GLP-1/ DPP-4pathway and glucolipid metabolism in adipose tissue of high-fat diet-induced obese mice.Methods:SPF grade aged 8 week male Kunming mice saperated to random 4groups: Contorl(,n=10),High Fat Diet(HFD,n=10),High fat diet+Sleeve Gastrectomy(HFD+SG,n=10)and High Fat Diet+Sham Operation(HFD+SO,n=10);Contorl group was given standard feed,other three group were given high-fat feed.After eight weeks of feeding,HFD+SO group and HFD+SG group were given surgical intervention.After surgery,they were given standard feed and continued to feed for four weeks.In the peroid of feeding after operation,weekly measured body weight of each mice.After operation,visceral adipose tissue samples were collected.The pathological changes of adipose tissue in each group were observed under light microscope by hematoxylin eosin(H&E)staining.The GLP-1,GLP-1R,DPP-4,NADPH oxidase 4(Nox-4)and proinflammatory cytokines(MCP-1,IL-1β,TNF-α)in adipose tissue in each group were analyzed by immunohistochemistry,real-time quantitative PCR.Results: H&E staining showed that the HFD group showed a large number of mononuclear cell infiltrative reaction and inflammatory changes in visceral adipose tissue.the SG intervention group significantly reduced the HFD-induced large number of mononuclear cell infiltrative reaction and inflammatory changes in visceral adipose tissue;the HFD group and the HFD+SO group significantly reduced the relative mRNA expression levels of GLP-1 and GLP-1R in adipose tissue(P<0.01);and their significantly increased the relative mRNA expression levels of DPP-4(P<0.01);the sleeve gastrectomy intervention(SG)group significantly increased the low expression of GLP-1 and GLP-1R mRNA levels in HFD-induced adipose tissue and significantly decreased the high expression of their DPP-4 mRNA levels(P<0.01).mRNA of Nox-4in HFD adipose tissue relative expression was significantly increased in the HFD adipose tissue,with a statistically significant difference compared with the Control group(P<0.001);the sleeve gastrectomy intervention(HFD+SG)group significantly reduced the expression level of Nox-4 in the adipose tissue,with a statistically significant difference compared with the HFD group(P<0.01).mRNA relative expression levels of these inflammatory factor indicators in the HFD adipose tissue were The mRNA expression levels of these inflammatory factors were significantly increased in the HFD adipose tissue,with statistically significant differences compared with the Control group(P<0.001),while the mRNA expression levels of MCP-1,IL-1βand TNF-ɑ in the adipose tissue were significantly decreased in the sleeve gastrectomy intervention(HFD+SG)group,with statistically significant differences compared with the HFD group(P<0.01).Conclusion: SG effectively regulate the abnormal expression of GLP-1/DPP-4 pathway,immune response,oxidative stress and pro-inflammatory cytokines in adipose tissue,thereby improving obesity induced inflammation of adipose.
