Objective: Focal cortical dysplasia(FCD)is an important cause of refractory epilepsy.At present,surgical resection is the main treatment.This study aims to explore the effects of SNAP-29 and STX-6 on clinicopathologic parameters of FCD.Methods:The gene expression profile of FCD GSE62019 was downloaded from the GEO database,and the differentially expressed genes were analyzed by the Limma software package of R software.Then functional enrichment was conducted by R software.The protein-protein interaction network was constructed by STRING,and its data was analyzed by Cytoscape module,which was verified by Gene Set Enrichment Analysis.Finally,enrichment analysis was carried out through Kyoto Encyclopedia of Genes and Genomes pathway and Gene Ontology analysis.The expression of SNAP-29 and STX-6in FCD patients was detected by immunohistochemistry.SPSS V.23.0 software was used to analyze and process the data.Spearman correlation analysis was used to analyze the relationship between the expression of SNAP-29 and STX-6.Independent sample t-test was used to analyze the relationship between FCD classification and the first onset age and course of disease.Chi-square test was not only used to analyze the relationship between FCD classification and the attack site,imaging results and attack form of patients,but also used to analyze the relationship between the expression of SNAP-29,STX-6 protein and the clinicopathological parameters of patients with FCD,and to compare the influence of the severity of FCD on Engel classification.Results: SNAP-29 and STX-6 are selected as research objects through bioinformatics analysis.In FCD,the age of first onset of type IIb is higher than type IIa(P=0.026),and the difference is statistically significant.The high expression of SNAP-29 and STX-6 is correlated with FCD Ⅱa(P<0.05)and FCD Ⅱb(P<0.05).High expression of STX-6 is associated with FCD Ⅱa and Ⅱb(P=0.005).The expression of SNAP-29 is significantly positively correlated with the expression of STX-6(P<0.01,r=0.515).FCD severity group is related to the postoperative efficacy of patients(P=0.004),and the postoperative efficacy of mild group was better than that of severe group.Conclusion: The expression of SNAP-29 and STX-6 is related to the classification of FCD,and SNAP-29 and STX-6may be used as potential biomarkers of FCD.
