| Objective: To screen the anti-asthma active site of Sorbus tianshanica Rupr based on BEAS-2B cell and to analyze their chemical composition.Methods: Different solvent extracts of Sorbus tianshanica Rupr were obtained by systematic solvent extraction method;the toxic effects of Sorbus tianshanica Rupr extracts on cells were investigated by MTT;an in vitro inflammation model was constructed using LPS-induced BEAS-2B cells,and ELISA was used to detect the levels of interleukin-1β and interleukin-8 to screen the anti-asthma active sites of Sorbus tianshanica Rupr;the composition analysis of the anti-asthma active extracts of Sorbus tianshanica Rupr was performed based on UPLC-QE-MS technique;the mechanism of anti-asthma action of Sorbus tianshanica Rupr was initially investigated based on network pharmacology and molecular docking technique.Results: 62.0 g petroleum ether extract,26.2 g chloroform extract,68.5 g ethyl acetate extract,134.6 g n-butanol extract and 904.0 g water extract were obtained from 5kg Sorbus tianshanica Rupr.MTT results showed that the concentrations of petroleum ether,chloroform,ethyl acetate,n-butanol and water extract were lower than 80、100、300、300 and 400 μg/m L,there was no toxicity to BEAS-2B cells.The results of ELISA showed that ethyl acetate extract,n-butanol extract and water extract could inhibit the secretion of IL-1β and IL-8 in LPS-induced BEAS-2B cells,and the inhibitory effect of ethyl acetate was more significant.The results of UPLC-QE-MS method indicated that the ethyl acetate extract of Sorbus tianshanica Rupr may contain flavonoids,terpenoids,phenols,phenylpropanoids,alkaloids,amino acid derivatives,organic acids and their derivatives,and the structures of 11 chemical components and their cleavage pathways were deduced from the fragment ions generated by secondary mass spectrometry.The results of network pharmacology showed that Sorbus tianshanica Rupr might exert its anti-asthma activity through signal pathways such as interleukin-17,tumor necrosis factor,Toll-like receptor,Th17 cell differentiation and HIF-1;The molecular docking results showed that the active ingredients could be well combined with the key targets.Conclusion: In this study,the anti-asthma activity of ethyl acetate,n-butanol and water extracts of Sorbus tianshanica Rupr was screened by constructing an in vitro BEAS-2B cell inflammation model,and the activity of ethyl acetate extract was more significant.The chemical composition of the ethyl acetate extract was analyzed based on liquid chromatography-mass spectrometry.The potential anti-asthma mechanism of Sorbus tianshanica Rupr initially explored by network pharmacology and molecular docking technology. |
