Background:Long-chain non-coding RNA(LncRNA)is a RNA with more than 200bp in length and almost no protein translation function in non-coding RNA.Recent studies have shown that LncRNA is widely involved in a variety of biological behavior abnormalities in tumorigenesis and development.The mechanisms of action of lncRNA and LincRNA,two different non-coding RNAs whose main differences lie in function,biological role and scope of influence,continue to be discovered in liver cancer.In this study,we mainly studied the mechanism of LincRNA00844 in hepatocellular carcinoma.Methods and Material:We utilized the GEPIA online analysis tool to investigate the expression pattern of Lincrna00844 in various tumor types.Furthermore,we examined the differential expression of Lincrna00844 in hepatocellular carcinoma(HCC)and its correlation with pathological staging.To accomplish this,we acquired RNAseq data(level 3)from the Cancer Genome Atlas(TCGA)database,comprising 158 Asian HCC tumor samples,along with their corresponding clinical information.Subsequently,we performed Kaplan-Meier survival analysis and time ROC analysis.Using the KEGG database,we retrieved genes associated with the Wnt/β-catenin pathway and demonstrated the relationship between these genes and Lincrna00844 using the R package pheatmap.To investigate the functional role of Lincrna00844,we employed lentivirus-mediated overexpression and knockdown techniques in cell culture.We then conducted a series of in vitro experiments,including CCK-8 cell proliferation assay,clone formation assay,scratch assay,and transwell assay,to assess the impact of altered Lincrna00844 expression on the proliferative and invasive capabilities of hepatocellular carcinoma cells.Additionally,we conducted Western blot experiments to explore the specific effects of Lincrna00844 on the classical Wnt/β-catenin pathway and its interrelationships.Result:The bioinformatics analysis showed that LincRNA00844 was lowly expressed in hepatocellular carcinoma compared with paraneoplastic tissues,and its expression was negatively correlated with the pathological stage and survival rate of hepatocellular carcinoma,and had some predictive ability.Cellular assays showed that LincRNA00844 had the ability to significantly inhibit the proliferation,migration and invasion of hepatocellular carcinoma cells in vitro,and that overexpression of LincRNA00844 induced inactivation of the wnt/β-catenin signaling pathway in hepatocellular carcinoma cells,while knockdown of LincRNA00844 caused activation of the Wnt/β-catenin signaling pathway in hepatocellular carcinoma cells.pathway activation.Conclusion:The level of LincRNA00844 in hepatocellular carcinoma tissues could be used as a potential prognostic molecule for patients with hepatocellular carcinoma or as a reference indicator to guide patients’ subsequent treatment after hepatectomy for hepatocellular carcinoma.The inhibitory effect of LincRNA00844 on the Wnt/β-catenin signaling pathway may be a new therapeutic target for patients with hepatocellular carcinoma.
