Objective:To explore the effects of EA on the behavior and morphology of slow transit constipation rats experimentally,we will reveal the mechanism by which EA can regulate Beclin-1,thereby regulating cell autophagy and protecting colonic tissue to function as a buffer against slow transit constipation and provide an experimental basis for the clinical treatment of slow transit constipation.Methods:The experiment was divided into two partsExperiment I: Effects of Electroacupuncture on gastrointestinal conduction function and colonic histopathology in rats with slow transit constipationA total of 24 SD male rats were selected and adaptively fed for 7d,and randomly divided into a blank group,a model group,a drug group,and an electroacupuncture group,with 6 rats in each group,and the blank group was left untreated,and the other three groups were all treated with compound diphenoxylate suspension by gavage and continuous gavage for 14 d to establish a rat model of slow transit constipation.After the successful modeling,the blank group and model group were single caged and not disposed.The drug group was given mosapride by gavage treatment,and the electroacupuncture group was given electroacupuncture intervention treatment for 14 d.The general status of the rats,the time of first black stool expulsion,fecal water content,and the small intestinal charcoal propulsion rate were observed;Hematoxylin eosin(HE)staining was performed to observe the pathological and morphological changes of colon tissues.Experiment 2: the protein expression levels of Beclin-1 were determined by immunohistochemistry in rat colon tissues.A total of 24 SD male rats were selected and adaptively fed for 7d,and randomly divided into blank,model,drug,and electroacupuncture groups wit h6 rats in each group,and the model preparation was the same experiment as the intervention method.The protein expression levels of Beclin-1 in rat colon tissues were determined by immunohistochemistry after the rats in each group were sacrificed.Results:1.Effects of Electroacupuncture on the general state of rats with slow transit constipation: during the modeling process,the mental state of rats in the model group versus the electroacupuncture group was better but eating was reduced,and the amount of feces was prolonged with the modeling time,which was gradually converted from the amount of loose stools to the amount of less and dry,and the emotional irritability appeared during the gavage process,the hair was dull and slightly yellow,and the color of urine was yellow.Rats in electroacupuncture group showed occasional aggressive behavior when acupuncture treatment was performed,and after treatment,the feces were slightly soft from dry and hard,the aggressive behavior was moderated,the hair resumed gloss,and the color taste of urine was normal.2.Effects of Electroacupuncture on the defecation of rats with slow transit constipation: compared with the blank group,the time of the first black stool discharge of rats in the model group was significantly longer(P < 0.01);Compared with the model group,the drug group,EA group rats treated for 14 d had a significantly shorter time to discharge of first grain black stool(P <0.01);There was no significant difference between the drug group a nd electroacupuncture group.Compared with the blank group,the fecal water content of rats in the model group was significantly decreased(P < 0.01);Compared with the model group,the fecal moisture content of the drug group significantly increased after 14 days of treatment(P<0.05),and the fecal moisture content of the electroacupuncture group rats significantly increased(P<0.01)..3.Small intestinal charcoal propulsion rate: compared with the blank group,the intestinal propulsion rate of rats in the model group was significantly decreased(P < 0.01);Compared with the model group,the small intestine char-acterization rate of rats in the drug group,EA group that had been treated for 14 d was significantly increased(P < 0.01).4.Effects of EA on the pathomorphology of colonic tissue in slow transit constipation rats: the colonic mucosal folds of rats in the blank group had an intact structure,and the number and arrangement of goblet cells were abundant.The colonic mucosal folds of the model rats were partially deficient,thinned,and showed reduced numbers of goblet cells,irregular arrangement,and intercellular edema.In the drug group,the structure of the colonic mucosal folds was more complete,and the number of goblet cells was moderate,their arrangement was relatively regular,and there was no edema among the cells;The colonic mucosa of EA rats was more intact,with more abundant goblet cells,relatively regular arrangement,and mild edema between the cells.5.Immunohistochemical method was used to detect the protein expression of Beclin-1 in colon tissue.The immunohistochemical results showed that compared with the blank group,the expression of Beclin-1 in colon tissue of model group rats increased,and the difference was statistically significant(P<0.01);Compared with the model group,the expression of Beclin-1 in the colon tissue of the drug group rats decreased,and the difference was statistically significant(P<0.05);Compared with the model group,the expression of Beclin-1 in the colon tissue of rats in the electroacupuncture group was significantly reduced,with a statistically significant difference(P<0.01).Conclusion:1.Electroacupuncture is able to improve the defecatory function of slow transit constipation rats to promote intestinal motility,and is able to effectively treat slow transit constipation.2.Electroacupuncture can inhibit Beclin-1 expression and regulate cell autophagy in the colonic tissue of slow transit constipation rats. |
