Objective:To capture data on brain-activation areas corresponding to acupuncture points on the left and the right sides of the Shenmen acupoints in healthy subjects with blood oxygenation level-dependent functional magnetic resonance imaging(Bold-f MRI),and to investigate whether acupoints between left and right sides have laterality of acupuncture effects, and to verify whether acupuncture methods such as Miuci recorded in ancient texts have clinical practical significance. We aim to provide an objective and visualized imaging basis for selecting acupoints for clinical treatments.Method:12 healthy subjects aged 24 to 28 years, all right-handers as inclusion criteria, were selected as the subjects for this trial. Each subject experienced two tests, respectively by acupuncture left and right Shenmen acupoints, with an interval of 14 days.Brain Bold-f MRI data of the subjects’ left(right)Shenmen acupuncture in the task state and the resting state with needle retention were acquired by Philips 3.0T dual-source and dual-gradient MRI(16 coils).Based on MRI convert,Matlab2022 a,Dpabi,Xjview96,Brainsetview2019,Spss V25.0 and Spm12.Then the data were formatted, preprocessed, statistically analyzed and outcome-presented.Finally,we got the different brain-activation areas of needling the left or right sides of the Shenmen acupoint by twisting and retaining needles, and the comparison between the left and right side of Shenmen acupoint, and the different Re Ho and ALFF values of needling the left and right sides by retaining needles. Finally,we concluded whether the brain activation of the left and right sides of the Shenmen acupoints in healthy individuals were directional by calculating and analyzing the data.Results:1.The positive activation areas in brain of acupuncture the right Shenmen acupoint: left middle occipital gyrus, left angular gyrus; The deactivation areas: right insula, right inferior frontal gyrus(triangular part),right rolandic operculum, right inferior frontal gyrus(opercular part),right superior temporal gyrus, right supramarginal gyrus, left inferior parietal gyrus, right supramarginal gyrus, right precentral gyrus.2.The positive activation areas in brain of acupuncture the left Shenmen acupoint:right caudate nucleus; The deactivation areas: left cerebellum 6,left hippocampus, right hippocampus, left superior temporal gyrus, right thalamus, left thalamus, left insula, left middle temporal gyrus, left supramarginal gyrus, left precentral gyrus, right supramarginal gyrus, left middle frontal gyrus, left caudate nucleus, right postcentral gyrus.3.Compared with acupuncture of the left Shenmen acupoint, the positive brainactivation areas in right acupoint: left caudate nucleus, left parietal lobe, left cingulate gyrus;the brain-deactivation areas: left cerebellum 8,left cerebellum crus 1,left fusiform gyrus, left superior temporal gyrus, right middle temporal gyrus, left middle temporal gyrus, left middle occipital gyrus, left supramarginal gyrus, left postcentral gyrus.4.Compared with retaining needles of the left Shenmen acupoint, the positive brain-activation areas in Re Ho values in right Shenmen acupoint: right cerebelum8,left cerebellum 8,left fusiform gyrus, left cerebellum 3,left Cerebellum 4-5,left superior frontal gyrus(orbital part),right middle temporal, left superior temporal gyrus, left superior frontal gyrus(medial),right middle cingulum gyrus;the braindeactivation areas: right superior frontal gyrus, right superior temporal gyrus, right postcentral gyrus, left inferior frontal gyrus(opercular part),left supramarginal, right cuneus, left middle occipital gyrus, left inferior parietal gyrus, right supramarginal gyrus, right precuneus, left precuneus.5.Compared with retaining needles of the left Shenmen acupoint, the positive brain-activation areas in ALFF values in right Shenmen acupoint: left Cerebellum 8,left cerebellum crus 2,left cerebellum crus1,left cerebellum 6,left inferior occipital gyrus, left middle temporal gyrus, right middle frontal gyrus;the brain-deactivation areas in ALFF values: left fusiform gyrus, left caudate nucleus, left middle cingulum gyrus, right precuneus.Conclusion:1.There are differences in brain-activation areas by needling the left or right side of the Shenmen acupoint in the same group.Brain-activation areas were mainly assembled on left brain when needling the left side of the Shenmen acupoint and vice versa. It indicates that the brain-activation areas of the left or right side of the Shenmen acupoint have laterality.2.The brain-deactivation areas mainly exist results from Shenmen acupoint. It suggests that neuronal activities in the brain-activation areas were predominantly inhibited by strong stimulation with acupuncture the Shenmen acupoint.3.There are significant differences in brain-activation areas results from contrasting brain-activation areas in task state and the resting state of acupuncture left or right Shenmen acupoint. It indicates that the treatment effects have laterality no matter in needling manipulation or retention of Shenmen acupoint.4.In both task state and resting state, there were significant differences in brainactivation areas between the left and right Shenmen acupoint groups, which were mainly concentrated in the left hemisphere.Further the activated areas of left hemisphere was more and broader.
